Objective: Resveratrol has received increasing attention as a phytochemical.In view of the fact that resveratrol can interact with metal ions such as zinc and copper in vitro,and in countries with plant-based foods,the incidence of iron deficiency and zinc deficiency is often higher than that of animal food countries.Therefore,this study explored the effects of resveratrol on trace elements in rats with normal and high-fat diets.Methods: 1.Solvent experiment: The absorption spectra of resveratrol with Fe,Cu,Zn and Mn in PBS were determined by ultraviolet visible light spectrophotometer.2.Animal experiments: SPF male Wistar rats(195.6±9.26g)were randomly divided into four groups,6 in each group: normal diet group(group C),high-fat diet group(group H),resveratrol intervention in normal diet group(C+R group)and resveratrol intervention in the high-fat diet group(H+R group).The resveratrol intervention dose was 100 mg/kg·bw,and the daily feed intake and weekly weight were recorded,and free drinking water.The intraperitoneal injection of glucose tolerance test(IPGTT)was performed at week 16.The 24-hour urine volume was collected in the metabolic cage at week 17.At the 18 th week,the rats were killed by decapitation.The organ coefficients were calculated and serum ALT,AST,blood glucose,blood lipids and whole blood glutathione peroxidase activity were measured.Liver tissue HE staining.The right femur of each group of rats was collected and frozen.Inductively Coupled Plasma Mass Spectrometry(ICP-MS)was used to determine the levels of five essential trace elements [iron(Fe)copper(Cu)zinc(Zn)selenium(Se)chromium(Cr)] and three toxic elements [lead(Pb)thallium(Tl)cadmium(Cd)] in feed,whole blood,femur and urine.Results:1.Resveratrol can interact with four elements of iron,copper,zinc and manganese in a solvent,and change its characteristic ultraviolet absorption spectrum.2.The average daily intake of rats in the high-fat group decreased,but the energy intake did not differ from the normal group.Rats in the high-fat group had higher body weight,visceral fat coefficient,area under the intra-peritoneal glucose tolerance curve,serum TC and LDL-C levels,and lower serum HDL-C levels.Resveratrol intervention in high-fat rats reduced body weight,TC,TG,and LDL-C,and increased serum HDL-C levels.3.High-fat diet induces fat vacuoles in the liver.Resveratrol has no significant effect on liver function(ALT and AST)and liver morphology,and it can alleviate high fat diet induced hepatic steatosis.4.The average daily intake of Fe,Cu,Zn,Se,Cr,Pb,Tl and Cd in the high-fat group was 46%,40%,59%,89%,67%,91%,67%,54% of the rats in the normal group.5.The Zn level in whole blood of the high-fat group was 52% of the normal group.Resveratrol reduced the levels of Cu and Tl in whole blood of the rats in the normal group by 64% and 77%,respectively,and increased the level of Se in the high-fat group by 17%.6.Resveratrol reduced the levels of Cr,Pb,Tl and Cd in the femur of the normal group by 85%,93%,50% and 50%,respectively,and the Fe level increased by 68%.Resveratrol increased the levels of Fe,Zn and Se in the femur of the high-fat group by 51%,17%,and 25%,respectively,while the levels of Cr,Pb,and Tl decreased by 96%,94%,and 43%,respectively.7.The level of Se excreted in the urine of the high-fat group was 48% of the normal group.Resveratrol increased the Cd excretion by 3.5 times in the normal group.Resveratrol reduced the levels of Cr,Tl,and Cd in the high-fat group by 60%,62%,and 83%,respectively.Conclusion:1.Resveratrol can interact with various metal elements in vitro solutions;2.In this experiment,the levels of intake of elements in normal and high-fat diet-fed rats were different,and the high-fat-fed rats were prone to zinc deficiency in whole blood;3.Resveratrol intervention had the same or different effects on the elements in the two dietary modes(normal and high fat).(1)The same: Resveratrol can increase the iron content in the femur of rats in two dietary modes,and reduce the accumulation of toxic elements lead and thallium.(2)Different: Resveratrol can reduce copper and thallium in whole blood of rats fed with normal diet;and increase the level of selenium in whole blood of rats fed with high fat diet.
