AIM and significance:The recurrence rate of advanced laryngeal cancer is high,and chemotherapy resistance is increasing,which is still the main cause of death of patients.So far,there is still no satisfactory solution,and it is urgent to explore new treatment from a new perspective.Previous studies showning that cucurbitin E has an inhibitory effect on tumor cells of multiple organs,but the inhibition of multi-drug resistance on laryngeal cancer tumor stem cells and its potential molecular mechanism are still unclear.The study aims to observe the inhibitory effect of Cucurbitacin E on laryngeal cancer stem cells,detect the expression of P-gp and ABCG2 protein on the surface of laryngeal cancer stem cells,and explore its molecular mechanism,so as to provide theoretical basis for clinical treatment of laryngeal cancer.Research method:1.To select Laryngeal cancer stem cells and culture2.To observe the effects of cucurbitacin E on the physiological activities(proliferation,invasion and migration)of laryngeal cancer stem cells,cucurbitacin E was cultured with laryngeal cancer stem cells3.To detect the expression of P-gp and ABCG2 protein in laryngeal cancer stem cells,and to explore its mechanism,cucurbitacin E was cultured with laryngeal cancer stem cellsResults:1.Cell proliferation experiment showed the proliferation ability and activity of laryngeal cancer stem cells after cucurbitacin E was decreased.2.Invasion-migration experiments showed that cucurbitacin E inhibited the invasion and migration of laryngeal cancer stem cells in varying degrees after co-culture.3.Western Blotting experiments showed that cucurbitacin E could decrease the expression of drug resistance-related proteins P-gp and ABCG2 in a dose-dependent mannerConclusion:Cucurbitacin inhibited proliferation,migration,and invasion of laryngeal CSCs.Further,we found that cucurbitacin decreased levels of ABCG2 and P-gp.This study provides a theoretical basis for guiding the clinical combination of drugs and improving the methods of chemotherapy in the treatment of laryngeal cancer,as well as for controlling the growth of tumors and improving the therapeutic effect of drugs.
