Objective:This study aims to observe the effect of nicorandil on IL-1? and IL-18 production in THP-1 macrophages as well as the regulatory roleofNLRP1 inflammasome in this process,and to explore the anti-inflammatory effect of nicorandil.Methods:THP-1 macrophages were cultured and divided into three groups for treatment: the blank control group,the LPS group,the LPS+ nicorandil group.The level of Il-1?and Il-18 in the supernatant was detected by ELISA,and the expression of NLRP1 mRNA and ASC mRNA was detected by qPCR.Results :The level of Il-1?(16.06±1.52 pg/ml)in LPS groupwere higher than that(3.50±1.60 pg/ml)in blank control group(p<0.01).The level of Il-1?(7.26±1.19 pg/ml)in nicorandil group were lower than that in LPS group(p<0.01).The level ofIl-18(25.62±1.87 pg/ml)in LPS group were higher than that(17.11±1.00pg/ml)in blank control group(p<0.01).The level of Il-18(13.58±1.34pg/ml)in nicorandil group were lower than that in LPS group(p<0.01).The expression level of NLRP1 mRNA(1.48 +0.21)in the LPS group was higher than that(1.01 + 0.13)in the blank group(p<0.05).The expression level of NLRP1mRNA(0.34±0.04)in the nicomandil group was lower than that in the LPS group(p<0.05).The expression level of ASC mRNA(1.61 + 0.16)in the LPS group was higher than that(1.06 + 0.35)in the blank group(p<0.05).The expression level of ASC mRNA(1.03 + 0.12)in the nicomandil group was lower than that in the LPS group(p<0.05).Conclusion:Nicomandil decreases IL-1? and IL-18 secretion via inhibition of NLRP1 inflammasome in THP-1 macrophages,possibly contributing to its anti-inflammatory effect.
