Objective:To study the effect of different vacuum pressure,needle type and number of needle passes on fine needle aspiration cytology by animal experiment.Methods: 24 healthy SD rats,male and female,with an average body weight of(200±13)g,were adaptively reared in the laboratory for one week before puncture.According to three different conditions of negative pressure(0,0.2,0.5 ml),needle type(21,22,23,25g)and number of times of lifting and inserting(5,10 times),24 different puncture methods can be generated correspondingly.24 rats were randomly selected for different puncture methods.During puncture,the puncture needle was inserted into the liver of rats with a fixed insertion depth of 1 cm and puncture times of 1 time.The puncture method corresponding to each rat was used for extraction and insertion.After smearing,HE staining was used to make the tablets.Under 400-fold light microscope,each slide was randomly selected from three fields to count the number of cells,and the average value was taken.The experiment was repeated three times and the average value was taken.Three-factor analysis of variance was used to compare the effects of different times of extraction and insertion,needle type and negative pressure on rat liver cell volume,and whether there was any interaction between the three.Results: The influence of needle type and vacuum pressure on cell volume was related to the times of extraction and insertion.When the number of times of lifting and inserting is5,the amount of cells obtained increases with the increase of puncture needle diameter and vacuum pressure(P <0.05).However,when the number of extractions and insertions is 10 and the needle type is 21 G and 22 G,the amount of cells obtained increases with the increase of vacuum pressure(P <0.05).When the needle type was 23 G and 25 G,the change of cell volume was not related to the increase of vacuum pressure(P >0.05).Under the same needle type and vacuum pressure,there are differences in the amount of cells in the samples taken at different times of extraction and insertion.When the needle type is 21 G,22 G,and any vacuum pressure,with the change of the number of times of extraction and insertion,there is a significant difference in the amount of hepatocytes obtained(P <0.001),and the amount of cells obtained with fewer times of puncture is more.When the needle type is 23 G and 25 G,the influence of the number of times of lifting and inserting on the cell volume is related to the change of vacuum pressure.In most cases,more cells can be obtained with fewer times of extraction and insertion.Conclusion: The number of times of extraction and insertion,needle type and vacuum pressure can affect the amount of cells obtained,and the three conditions affect each other.When the number of times of lifting and inserting is less,the diameter of puncture needle is larger and the vacuum pressure value is higher,the number of cells obtained is more.
