Objective: To observe the theraputic of human umbilical cord mesenchymal stem cells(hUC-MSCs)treatment to the microvascular density and neurogenesis in APPswe/PSNE1 double transgenicmice.Methods:HUC-MSCs were injected to 3 and 12 months APPswe/PSEN1 double transgenicmice(n=10),the same amount of PBS were injected the control groups(n=10).After the second,fourth and sixth injection in each group,use immunohistochemical methods to observe microvascular change and NeuN in the brain.Results: APPswe/PSEN1 double transgenic mice microvascular density decreased with the increase of age.The microvascular density were significantly different between hUCMSCs group and PBS group in 3-month-old group(P < 0.05);The microvascular density were significantly different between hUC-MSCs group and PBS group in 12-month-old group(P < 0.05).Neuron specific nuclear protein(NeuN)positive cells in dentate gyrus and subgranular area of hippocampus were significantly different between 12-months hUC-MSCs group and PBS group(P < 0.05).Conclusion:With the increase of age,the microvascular density in the mice brain gradually decreased,which indicated that microvascular changes were involved in the occurrence and development of AD in APPswe/PSEN1 transgenic mice.Human umbilical cord mesenchymal stem cell therapy could prevent and improve the microvascular change in the brain,and promote the nerve regeneration of dentate gyrus in the hippocampus of APPswe/PSEN1 transgenic mice.
