Object: To observe the abnormal changes of pericytes in the blood-brain barrier opening after epilepticus,to explore the role of pericytes in the acute blood-brain barrier opening and chronic blood-brain barrier repair after epilepticus,and to treat the status of epilepsy.Scientific research provides new ideas and methods.Method: The rat model of seizure induced by lithium chloride-pilocarpine was prepared.The degree of blood-brain barrier damage in hippocampus was measured by Evans blue method and dry-wet weight method at 2h,24 h,3d,7d,14 d and 21 d after epilepticus.The ultrastructural changes of pericytes and blood-brain barrier were observed by electron microscopy.The expression of pdgfrβ in hippocampus was detected by Western Blot.The immunohistochemistry and immunofluorescence were used to semi-quantitatively analyze Pdgfrβ.Distribution and expression in the hippocampus,observing the role of pericytes in the destruction and recovery of epileptic seizures in the acute and chronic phase.Results: 1.The blood-brain barrier is destroyed after the status of epilepsy,and the permeability is enhanced.The damage is most serious at 24 h after the attack,and gradually returns to normal after 3d and 2 days.2.The results of transmission electron microscopy show that the microvascular pericytes and their basement membranes are supermicroscopic after SE attack.The structure was changed.3.The results of Western blot and immunofluorescence showed that the expression level of Pdgfrβ was significantly higher than that of the control group 24 h after SE attack(P<0.05),and gradually became stable after 3 d and later.4.The results of immunofluorescence showed Pdgfrβ along The distribution and distribution of Pdgfrβ at 24 h after SE onset were significantly higher than those in the control group(P<0.05),and gradually stabilized at 3 d and later.Conclusion: The blood-brain barrier was the most severe after 24 hours of epileptic seizures,and gradually returned to normal after 3 days and later.Pericytes played an important role in the mechanism of destruction and subacute recovery in acute phase.It can play an important role in clinical treatment of early brain damage in SE.
