| Objective:The cold atmosphere plasma(CAP)is an approximately electrically neutral ionized gas composed of positively and negatively charged particles and various neutral particles.In some malignant tumors can participate in the anti-tumor process through a variety of mechanisms.Nasopharyngeal carcinoma is a common malignant tumor of head and neck with a hidden pathogenesis and poor prognosis.The aim of this study was to observe the effects of cold atmosphere plasma treatment on the proliferation,migration and apoptosis of nasopharyngeal carcinoma cells,in order to find more effective techniques and treatments for improving the prognosis of nasopharyngeal carcinoma.Methods:The CNE002 cell were treated with CAP for 0s,15 s,30s and 60 s respe ctively.The morphology of cells after CAP treatment at different lengths was obs erved under an inverted microscope,CCK-8 method was used to detect the cell pr oliferation ability.The cell migration ability was detected by scratch test and trans well method.Flow cytometry and Western blot were used to determine apoptosis a nd expression of apoptosis-related proteins.After loading the DCFH-DA probe into the cells,the changes in the intracellular reactive oxygen species were observed under a fluorescence microscope.Data were analyzed using one-way analysis of va riance,and statistical analysis was performed using SPSS 22.0 statistical analysis s oftware.Results:1.Under the inverted microscope,it was found that the CAP treatment made the cell volume smaller and the shape became round.When the treatment ti me reached 60 s,the adhesion ability of some adherent cells to the substrate decre ased,and it floated in a cluster.2.CCK-8 results showed that the cell proliferatio n ability was significantly inhibited after CAP treatment(P<0.001),and this inhibi tion was related to the treatment time.3.Scratch test and transwell test results sh owed that CAP treatment significantly inhibited cell migration ability(P<0.05),an d this inhibition was also related to treatment time.4.The results of flow cytomet ry and Western blot showed that CAP could promote apoptosis(P<0.05).When th e treatment dose was 30 s,60s,the expression of proapoptotic protein Bax was significantly up-regulated(P<0.05).5.The results of fluorescence microscopy showed that with the increase of treatment dose,the intracellular ROS content increased,suggesting that CAP can increase the intracellular reactive oxygen species.Conclusion:Cold atmosphere plasma has the ability to inhibit proliferation,migrat ion and promote apoptosis of nasopharyngeal carcinoma,and it can increase the c ontent of reactive oxygen species in nasopharyngeal carcinoma cells. |
