Objective: SIRT6 belongs to the family of Sirtuins and is a homolog of the yeast silencing information regulator Sir2.The research of SIRT6 has been extended to many types of diseases,and the regulation of SIRT6 on various life activities has been gradually clarified.Accompanied by these studies,there is a rapid development of SIRT6 related literatures.However,the scientometric information characteristics of SIRT6 researches are still unknown.In tumors,SIRT6 is a double-edged sword that can act as both a tumor suppressor and a tumor promoter.However,no studies have revealed the biological function of SIRT6 on renal cell cancer and its association with the prognosis of patients with renal cancer.Therefore,we will use this as an opportunity to explore these two aspects.In the part of scientometric analysis of SIRT6 studies(Part 1),we will use a series of scientometric analysis tools to analyze the bibliometric information of SIRT6 and explore the scientometric characteristics of SIRT6 studies.In the research of the role of SIRT6 ob renal cell carcinoma(Part 2),we will identify the role of SIRT6 on the proliferation,migration and invasion of renal cancer cells as well as its role in chemosensitivity to cisplatin.Finally,we will lay a theoretical foundation for targeted therapy of renal cell cancer.Methods: In the scientometric analysis part of the SIRT6 studies(Part 1),this study retrieved the core collection of the web of science database of 2000.01.01-2018.04.28 for the research article of SIRT6 as the research sample.There are 441 articles included in the literature set and they are downloaded and stored as TXT format.Bicomb software was used to extract high frequency words and to construct a binary matrix as well as a co-word matrix.We use Gcluto for double cluster analysis.We use EXCEL to build the strategic coordinates and use Citespace software for co-citation analysis,Vosviewer for journal and density analysis.In the part of the role of SIRT6 on renal cell carcinoma(Part 2),we has analyzed the transcription expression difference of SIRT6 between renal cell carcinoma and normal tissues by ULCAN,a online bioinformatic analysis tool.And explored ralativity betweenthe transcription expression level of SIRT6 and the pathological grading as well as the prognosis of renal cell carcinoma.In the 786-O,769-P cell line of renal cell carcinoma,SIRT6 was knocked down by siRNA,and the effects of SIRT6 on proliferation,migration and invasion of renal cancer cells as well as chemosensitivity were examined by colony formation assay,CCK8 assay,cell scratch assay and transwell assay accordingly.The knockdown efficiency of siRNA and its effect on downstream proteins were identified by Western blotting assay.Results: In the scientometric analysis part of the SIRT6 studies(Part 1),we used the co-word analysis method to get eight research hotspots of SIRT6 studies and the changes of SIRT6 hot terms over time.We constructed the hot-spot strategic coordinates to judge the research prospects and connections among these various hotspots.Then,another eight knowledge bases of SIRT6 were obtained by constructing the co-citation network and clustering analysis of the network.The most important nodes in the SIRT6 co-citation network were identified.At the same time,the distribution of subjects and journals of SIRT6 were visually analyzed.In part of the role of SIRT6 on renal cell carcinoma(Part 2),bioinformatics analysis showed that SIRT6 is overexpressed in renal cell carcinoma,and its expression level is related to tumor pathology grading and prognosis.Through CCK8 experiments and colony formation experiments,it was found that SIRT6 has the ability to promote the proliferation of renal cancer cells.Through cell migration experiments and transwell experiments,it was found that SIRT6 can promote the ability of migration and invasion of renal cancer cells.Knocking down SIRT6 can increase the sensitivity of cisplatin chemotherapy to Cisplatin.The down-regulation of NF-kB,cyclinD1 protein expression,up-regulation of Beclin1 protein expression were also found when SIRT6 was knocked down.Conclusion: In the scientometric analysis part of the SIRT6 studies(Part 1),the research draws the following conclusions:(1)SIRT6's regulation of lifespan,chromatin,DNA damage repair,and metabolism make up the most important SIRT6 research basis from the past ten years.(2)SIRT6 studies has concentrated on the effects of its role on tumors and shown promising trends in neural diseases.(3)Research by Motoslavsky represents a trigger research in the whole SIRT6 research co-citation network,and the work byKawahara and Kim are vital in the knowledge inheritance and transmission of SIRT6 research co-citation network.In the part of the role of SIRT6 on renal cell carcinoma(Part 2),the study concluded that:(1)SIRT6 is highly expressed in renal cell carcinoma,and its expression level is related to tumor pathology grading and prognosis,and patients with high expression have poor prognosis.(2)SIRT6 promotes the ability of proliferation,migration and invasion of renal cancer cells.And knocking down SIRT6 can increase the chemotherapy sensitivity of renal cancer cells to Cisplatin.(3)In the renal cell carcinoma 769-P cell line,SIRT6 can promote the protein expression of NF-kB,cyclin D1,and inhibit the expression of Beclin-1.
