Background: Rapid development of oral and maxillofacial surgery nowadays,jaw defect has become a problem that needs urgent solution.Through experiments,it was found that adipose stem cells with multi-differentiation ability have high stable value-added and lower mortality rate in vitro culture,and they have a wide range of sources and easier separation and culture characteristics.There is evidence that wolfberry can not only regulate the differentiation of adipose stem cells into osteoblasts,but also accelerate the formation and precipitation of bone matrix proteins.And promote the synthesis and precipitation of bone matrix proteins.It has been reported that the erbium ion promotes the optimal concentration of adipose-derived stem cells for osteogenic differentiation,and the effects of indole-doped nano-hydroxyapatite on the osteogenic efficiency of adipose-derived stem cells have not been reported in detail.Objective: Experimental grouping: group A: general cell culture medium + strontium-doped nano-hydroxyapatite scaffold(ASCs/n-SrHAP)seeded with adipose-derived stem cells;group B: DMEM osteogenic conditioned medium +(ASCs/ n-HAP)group;group C;ordinary cell culture +ASCs/n-HAP treated with adipose-derived stem cells Phosphatase activity,cell proliferation,expression of osteoblast-related proteins,and definite time conditions on the effect of strontium-modified nano-hydroxyapatite on the osteogenic efficiency of ADSCs.Experimental method: 1.Adipose stem cells isolation,culture and identification;2.Preparation of a fixed concentration of strontium-doped hydroxyapatite scaffold;3.The ASCs / n-SrHAP scaffolds were formed by the combination of ASCs / n-SrHAP materials and the control group:(1)DMEM osteogenic conditioned medium + ASCs / n-HAP scaffolds;(2)ordinary cell culture medium + ASCs / n-HAP scaffold;4.To detect the cell viability of ASCs at different time points,and to clarify the effect of time-dependent conditions on the osteogenic differentiation of ADSCs treated with strontium-doped nano-hydroxyapatite.Experimental results: Group A: ASCs/n-SrHAP scaffold materials General cell culture fluid and group B: DMEM osteogenic conditioned medium + ASCs/n-HAP scaffold group can detect alkaline phosphatase;A,B group alkaline phosphatase content No statistically significant difference and all greater than group C: normal cell culture fluid + ASCs/n-HAP scaffold.Conclusions: Strontium-doped nano-hydroxyapatite can promote the osteoblastic differentiation of ADSCs.The osteoblastic effect of ADSCs increased rapidly from day 4 to day 7,and the osteoblastic effect slowed down from the 7th day to the 10 th day.From the 10 th day to the 18 th,the osteogenic potential basically remained unchanged.
