The accumulation of active ingredients in medicinal plants was regulated by such factors as genotype and environment.Tetrastigma Hemsleyanum Diels et Gilg from different producing areas in northern Fujian was selected as the research object,and rutin was used as the evaluation index of flavonoid active ingredients.The fingerprint was established in eight different producing areas in northern Fujian.The effects of different origin factors on the content of rutin,an active component of Tetrastigma Hemsleyanum Diels et Gilg flavonoids were studied.The optimal extraction condition of Rutin was determined by four single factors: methanol concentration,ultrasonic power,extraction time and olid-liquid ratio.HPLC was used to determine the Rutin concentrations in the Tetrastigma Hemsleyanum Diels et Gilg.The factors of different extractors,chromatographic column,Extraction method and origin were discussed and determined the fingerprint.It provided the experimental basis for the determination of the content of Rutin and the formulation of the fingerprint of Tetrastigma Hemsleyanum Diels in different producing areas in northern Fujian.Main results were as follows: 1?Study on extraction of Rutin from Tetrastigma Hemsleyanum Diels et Gilg.The results showed that 60 % methanol concentration,20 min extraction time,324 W ultrasonic power,and 1:50 g/mL olid-liquid ratio,it was the best technical conditions for the extraction of Rutin in Tetrastigma Hemsleyanum Diels et Gilg.Determined the content of Rutin in Tetrastigma Hemsleyanum Diels et Gilg by HPLC.The method was used by agilent Allsphere ODS-?(250 × 4.6 mm,5 ?m)column,0.1 % aqueous phosphate solution(16:84),360 nm Wavelength,40 °C temperature,1 mL/min Flow rate,10 ?L sample volume.The concentration of rutin was in the range of 5 ?g / mL to 100 ?g / mL,and the regression equation between peak area(Y)and rutin concentration(X)was Y = 16.08532 × X-19.01623,R=0.9992;The sample calibration recovery rate was 95.00 %-99.56 %,the average markup recovery rate was 98.30 %,and RSD = 1.53 %(N = 6).Experimental results showed that the content of Rutin in Tetrastigma Hemsleyanum Diels et Gilg was 0.36-3.28 mg/g in different sample.Rutin content order in Tetrastigma Hemsleyanum Diels et Gilg from 8 producing areas: Huaqiao Town Songxi County>Jinhang Town Shaowu City>Shangmei Township Wuyi City>Shiqian Town Guangze County>Waitun Town Zhenghe County>Yangkou Town Shunchang County >Fuling Town Pucheng County>Shaowu Nanwuyi Pharmaceutical Park.2?Taking the three major technical bottlenecks as the background of the development of the industrialization of Tetrastigma Hemsleyanum Diels et Gilg in northern Fujian,HPLC was used to analyze the index components of the Tetrastigma Hemsleyanum Diels et Gilg.The fingerprint analysis method was established and its analyzed the index components of the Tetrastigma Hemsleyanum Diels et Gilg by HPLC.The experimental results showed that the active components of Tetrastigma Hemsleyanum Diels et Gilg were done.It was extracted by using methanol 40 mL,20min extraction time,and 324 W ultrasonic power.The method was used by agilent5 TC-C18(2)(150 × 4.6 mm,5 ?m)column,0.1 % aqueous phosphate solution(16:84),360 nm Wavelength,40 °C temperature,1 mL/min Flow rate,10 ?L sample volume.The results showed that the similarity of the content measured in 8 batches of Tetrastigma Hemsleyanum Diels et Gilg was between 0.815 and 0.926,it indicated that the fingerprint similarity of the Tetrastigma Hemsleyanum Diels et Gilg in northern Fujian was good.It can provide a certain basis for the identification and quality control standards of Tetrastigma Hemsleyanum Diels et Gilg in northern Fujian Province.
