As one of the malignant tumors that endanger women's health,breast cancer is related with many regulatory molecules and signaling pathways,and its development process needs further study.MicroRNA is one kind of non-coding RNA,which has been shown to play an important role in the diagnosis and treatment of many diseases.This study aims to identify the main target of potassium piperate(GBK)by analysing miRNA expression changes under GBK treatment.The results showed that GBK inhibited the expression of tumor suppressor gene miR-31,and in turn inhibited cell migration through downregulating the expression of cell migration-related proteins RhoA,WAVE3 and SATB2.In a drug combination study with cisplatin and etoposide,it was found that the drug combination strategy also promoted the up-regulation of miR-31 expression,as well as miR-31 target gene SATB2.The mouse tumor xenograft model confirmed that GBK combined with chemotherapy drugs can significantly inhibit tumor growth.These results indicate that GBK targets miR-31 and inhibits the migration of breast cancer cells by promoting its up-regulation.GBK mediated-drug combination strategy can enhance the suppression effect of chemotherapy drugs.In conclusion,GBK is expected to be developed as an auxiliary drug for clinical treatment of breast cancer.
