ObjectiveTo investigate the protective effect of folic acid on lung ischemia reperfusion injury in rats.Methods30 healthy adult male SD rats were randomly divided into group C(sham operation group),group I/R(ischemia reperfusion group),FA group(ischemia reperfusion after folic acid feeding group),FA group is further divided into FA1,FA2 and FA3 groups,which was given Folic acid at 5 mg/d,10 mg/d and 20 mg/d for one week before the operation.Dissociate the hilus pulmonis of group C but not clamped,while clamped for 30min and reperfusion for 2h in group I/R and group FA.After surgery,HE staining to observe the pathological changes of lung tissue in each group of rats.The levels of serum interleukin-8(IL-8)and tumor necrosis factor alpha(TNF-?)were determined by enzyme-linked immunosorbent assay(ELISA).Results1.Pathological changes and damage score in lung tissue:Observed by HE staining,the structure of lung tissue in group C was almost complete,and the alveolar septum was almost normal;No obvious exudate was found in the alveoli,and no obvious edema and inflammation were observed in the pulmonary interstitial.In the group I/R,alveolar and interstitial diffusely hemorrhage,edema,partial alveolar rupture to fusion,the alveolar septum thickening.The lung injury in the group FA was less than that in the group I/R,and the degree of injury in the groups FA1,FA2,and FA3 decreased in turn.Evaluate the degree of lung injury in each group with the Smith score and the results showed that,compared with group C,the injury severity of lung in group I/R was significantly more serious;compared with group I/R,the severity in the three groups of FA was alleviated,which was reduced in turn in FA1,FA2,and FA3.The difference was statistically significant(P<0.05).2.IL-8 and TNF-a:Compared with group C,the expressions of IL-8 and TNF-?in group I/R and the three FA groups were higher,and the difference was statistically significant(P<0.05).Compared with group I/R,the level of IL-8 and TNF-? in the FA groups were significantly lower,and decreased with the increase of folic acid dose,the difference was statistically significant(P<0.05).ConclusionFolic acid has a protective effect on lung ischemia-reperfusion injury in rats,and this effect is enhanced with the increase of folic acid dose within a certain range.
