Background: Diabetic nephropathy is an important cause of end-stage renal disease.Traditional Chinese medicine Panax notoginseng can improve the diabetic nephropathy,but its main effective components and the mechanism have not been elucidated.Notoginseng Fc is a new monomer of Panax notoginseng with anti-platelet aggregation effect.The study on the protective effect and molecular mechanism of Notoginseng Fc may provide new ideas and methods for the prevention and treatment of diabetic nephropathy.Objectives: Study the protective effect of Notoginseng Fc on the kidney of type 2 diabetes mellitus(db/db)mice,explore the protective effect and molecular mechanism of Notoginseng Fc and provide new ideas and methods for the prevention and treatment of diabetic nephropathy.Methods: This experiment gavaged db/db mice by Notoginseng Fc for 8 weeks and then detected urinary protein and other renal function index,observed the renal pathological changes,apoptosis of podocytes,expression of podocyte adhesion related molecules,such as,vWF,ADAMTS13,NLRP3 inflammasome,to study the renal protective effect of Notoginseng Fc on diabetic db/db mice.Results: 1)Notoginseng Fc can reduce proteinuria,podocyte injury,improve the renal function of db/db mice;2)Notoginseng Fc,can downregulate the expression of vWF,upregulate the expression of ADAMTS13,relieve the microcirculation disorder,decrease NLRP3 inflammasome activity,inhibiting the inflammatory reaction,delay the progress of renal disease,has the protective effect of diabetic nephropathy.Conclusion: Notoginseng Fc has protective effect on kidney of db/db mice by regulating microcirculation disorder,inhibiting inflammatory and alleviating podocyte injury.
