Objective: To determine the expression levels of NUMB in neuroblastoma cell lines,and the relation between NUMB and MYCN.Methods: Four cell lines: SK-N-BE(2),IMR-32,SK-N-SH and SH-SY5 Y were selected.qRT-PCR was performed to detect MYCN and NUMB expression in neuroblastoma cells.Western blot was used to detect the NUMB relative protein expression.Results: The high expression of MYCN and NUMB relative mRNA in SK-N-BE(2)and IMR-32,on the contrary the low expression of them in SK-N-SH and SH-SY5 Y.In protein level NUMB was highly expressed in SK-N-SH and SH-SY5 Y and low in SK-N-BE(2)and IMR-32.Conclusions: In the MYCN amplificated cell lines,expression of NUMB relative mRNA in SK-N-BE(2)and IMR-32 is high,in protein level low.Inversely,in the MYCN non-amplificated cell lines,expression of NUMB relative mRNA in SK-N-BE(2)and IMR-32 is low,in protein level high.NUMB may act as a cancer suppressor gene in neuroblastoma. |
