Objective:The aim of this study was mainly to discuss the effect and mechanism of miRNA-145 on the migration of human periodontal ligament fibroblastsMaterials and methods:The human periodontal ligament fibroblasts were cultured by the method of enzyme digestion in vitro and gone down to the future generation.The cells were divided into two groups of the control group and the experimental group of transfecting miRNA-145 mimics.The cells of the experimental group were transfected with 50 ng/ml miRNA-145.The proteins of the two groups were extracted and the related changes of expression level of ROCK 1 were analyzed by Western Blot analysis.Used the scratch test to measure the relevant changes of the distances between the scratched cells of the two groups and chose the points in time of 0 hour?24 hours?48 hours?72 hours after scratching and measured the distances between scratched cells and computed the means and stated the effects of miRNA-145 on the migration of the human periodontal ligament fibroblastsResults:Compared with the control group,the expression level of the target protein ROCK1 was significantly lower in the experimental group(p<0.05)after the cells were transfected with miRNA-145 mimics;and the distance between the scratched cells of the experimental group was significantly higher than that of the control group at 24 hours and 48 hours after transfecting(p<0.05)Conclusions:miRNA-145 could inhibit the migration of human periodontal ligament fibroblasts via down-regulation the expression of ROCK 1.The study can help us understand the mechanism of miRNA-145 in the progress of tooth movement.
