| Anti-Mullerian hormone(AMH)is a sex hormone in women.It is the only expression in the ovarian mid-anterior follicles and sinusoidal follicles.It is the biomarker with the highest predictive value for ovarian reserve function and ovarian response found in the latest clinical research.The traditional AMH detection system relies on foreign biological reagents and instruments companies to provide,and requires professional technicians to operate large-scale instruments and equipment to complete.Only a small number of top three hospitals or research institutes can carry out,the penetration rate is relatively low,and the testing cost is high,which seriously restrict the promotion of AMH detection.Therefore,the modern reproductive health market urgently needs a rapid,efficient,stable and sensitive artificial AMH detection system,and incorporates AMH detection into routine clinical tests to provide a full range of reproductive health protection for infertility patients.Blu-ray Disc can not only be used as a storage medium for ultra-clear movies,but also can replace traditional silicon wafers as a base material for biochemical reactions due to its excellent optical properties and nano-level surface flatness,and combined with high-precision optical reading,the servo control system and opto-mechatronics technology of Blu-ray disc drives,which can be used to develop new high-throughput analytical sensing devices.Based on the principle of Blu-ray Disc/optical drive reading,this paper independently develops a scanning imaging system based on Blu-ray disc in the Windows environment by collecting the light intensity signal of the optical head inside the optical drive,and designs and integrates a high-throughput biomolecular lattice quantitative detector with the advantages of full-disc imaging and high-throughput detection.In this paper,we mainly studies the feasibility analysis experiment,biomolecule and AMH quantitative detection experiment based on Blu-ray disc scanning imaging system.The Blu-ray disc is used as the reaction substrate,and the Blu-ray disc is used as the analysis device.Since the bioreactor array on the surface of the disc interferes with the normal reflection of the laser beam of the optical disc,the light intensity signal received by the photodetector in the optical head is lowered.The self-developed Blu-ray disc scanning imaging system collects the light intensity signal of the photodetector in the optical disc during the scanning process of the optical disc,and performs data processing and imaging analysis to realize quantitative detection of biomolecules.The main research contents of this paper are as follows:1.Based on the Blu-ray disc scanning imaging system,the Blu-ray disc drive,data acquisition card and portable industrial computer are designed and integrated with a high-throughput biomolecular dot matrix quantitative detector,which can be used for rapid quantitative detection on site,the resolution can reach 100 μm,the maximum detection throughput is 10,000 samples.2.The UV/Ozone treated Blu-ray disc surface was printed with inkjet dot matrix,and the linear relationship between dot matrix size and gray level and pixel value was obtained through system imaging analysis,which verified the high throughput and feasibility of the system.Bio-disc was prepared by the combination of biotin-streptavidin reaction system and silver-enhanced signal amplification method with polydimethylsiloxane(PDMS)microchannel technology.The method of Blu-ray disc scanning imaging system was used to achieve quantitative detection of different concentrations of streptavidin.3.Using the double antibody sandwich immunoreaction as the detection principle,the quantitative detection of AMH was realized by the imaging system method.The detection range was 0.05-25 ng/mL and the limit of detection was 0.05 ng/mL,which met the requirements of clinical diagnosis.Comparing AMH with several other sex hormones,the experimental results show that AMH has good specificity.Comparing with the conventional large-scale instrument chemiluminescence immunoassay method,the experimental results verify the accuracy and reliability of the method for AMH detection.. |
