Objective:To investigate the correlation between the distribution of common polymorphic loci of cytochrome cyp2c19 gene and cerebral ischemic stroke among Chinese Tibetans from Qinghai Province.Methods:Tibetan residents from the inpatient department,outpatient department and physical examination center of Qinghai Provincial People’s Hospital were included and selected continuously,including 52 patients with cerebral ischemic stroke and 60 healthy people as contrasts.All research subjects were selected according to the relevant inclusion and exclusion criteria.The venous blood DNA of all subjects was extracted,and real-time fluorescence quantitative PCR(q-PCR)method was used to detect the CYP2C19 common polymorphic site.The chi-square test was used to conduct a statistical analysis on the CYP2C19 genotype and distribution of frequency of occurrence of allele in stroke patients and the control group.Multivariate logistic regression was used to further analyze the correlation between different CYP2C19 genotypes and cerebral ischemic stroke in Tibetan population.Results:1.The stroke group was found to have a markedly higher constitute ratio of hypertension(33.3%vs.76.9%,χ~2=21.281,P<0.0001),ischemic heart disease(15.0%vs.30.8%,χ~2=3.995,P=0.046)and a higher ratio of systolic blood pressure[(123.57±22.07)mmHg vs.(139.63±17.63)mmHg,t=-4.212,P<0.0001]to diastolic blood pressure[(78.23±13.84)mmHg vs.(85.67±14.53)mmHg,t=-2.772,P=0.007]than the control group;There was no significant difference between the two groups in terms of composition ratio of sex,smoking history and diabetes and age,TC,TG,HDL-C,and LDL-C.2.The AA genotype of locus of rs4244285 of CYP2C19 gene was markedly higher in the ischemic stroke group than in the control group(χ~2=6.142,P=0.013).The GG genotype was common in both groups.The frequency of occurrence of A allele in the ischemic stroke group was markedly higher than that in the control group(χ~2=6.798,P=0.009).Multivariate logistic regression of risk factors for ischemic stroke suggested that AA genotype was an independent risk factor for cerebral ischemic stroke(OR=6.043,95%CI=1.034~35.310,P=0.046).3.In the overall study population,the frequency of occurrence of CYP2C19*2 allele was 29.0%,the frequency of occurrence of*3 allele was4%,and the frequency of occurrence of*17 allele was 4%.Conclusion:1.The CYP2C19*2*2 homozygous mutant genotype of rs4244285locus of CYP2C19 may be an independent risk factor for cerebral ischemic stroke in Tibetan population.2.Gender may not be the main factor that influences gene polymorphism of CYP2C19.
