Objective To investigate the difference between the expression of Serine/threonine kinase 33(STK33)in thoracic esophageal squamous cell carcinoma(ESCC)and adjacent normal tissue.To explore the relationship between STK33 expression in thoracic ESCC tissues and general clinicopathological data and prognosis.Further to learn the possible mechanism and potential clinical value of STK33 expression in thoracic ESCC.Methods Collecting tumor hospital of ningxia medical university general hospital between January 2008 and December 2012,a total of 358 patients with surgical treatment of ESCC,collect the clinical,pathological and follow-up data,after the inclusion criteria,exclusion criteria delete some patients,to the follow-up of 218 patients with thoracic segment ESCC,from 124 patients with complete follow-up data of 80 patients were randomly selected as a research object,draw on its tissue adjacent to carcinoma tissue and corresponding wax block,by HE staining to observe the pathological characteristics,immunohistochemical SP method to detect STK33 expression of the situation,To analyze the relationship between STK33 expression,clinicopathological indicators and prognosis.SPSS17.0 software was used for statistical analysis of relevant data,and P < 0.05 was considered as specific statistical difference.Results STK33 expression in ESCC of thoracic segment was significantly higher than that in adjacent normal tissues(5%,P < 0.05).STK33 positive expression rate was 60%(18/30)in keratinized squamous cell carcinoma,and 100%(50/50)in non-keratinized squamous cell carcinoma,with statistically significant difference(P < 0.05).The positiveexpression rate of STK33 in T3/T4(95.5%,42/44)was significantly higher than that in T1/T2(72.2%,26/36,P<0.05).Positive expression rate of STK33 in ? period was 88.9%(24/27),significantly higher than that in ? / ? expression in 34/53(64.2%,P < 0.05);Positive expression of STK33 in thoracic ESCC pathological and histological differentiation of different types of(Angle type squamous cell carcinomas,Angle type squamous carcinoma),infiltration depth(T1 / T2,T3 / T4)and pathological staging(?/?,?)differences between statistically significant(P < 0.05);The 5-year survival rate of STK33 positive expression group was 32.4%,significantly lower than that of the negative expression group(66.7%,P <0.05).There were statistically significant differences in tumor size,tumor invasion depth,lymph node metastasis,histopathological type,postoperative pathological stage,postoperative adjuvant therapy,and STK33 expression among the risk factors affecting the prognosis of patients by univariate log-rank analysis(P < 0.05).Multivariate analysis showed that tumor size and protein kinase STK33 expression were independent risk factors for prognosis of thoracic ESCC patients(P < 0.05).Conclusions STK33 positive expression was significantly increased than that in normal tissue,which means STK33 may play an important role in the tumorigenesis of thoracic ESCC.While there were closely relationships between STK33 obviously higher expression and poorer differentiation,deeper invasion and higher pathological stage,STK33 possibly participates in the invasion and metastasis in ESCC.Markedly increased expression of STK33 was an independent predictor of poor prognosis in ESCC.There is a potential clinical value that STK33 protein overexpression may be served as an early diagnostic marker and prognostic predictor in ESCC.
