Objective:To investigate the expression of GPR120(G protein-coupled receptor 120,GPR120)in breast cancer and observe the effect of GPR120 on the proliferation,migration and invasion of breast cancer cells.Methods:1.Breast cancer tissues and adjacent normal tissue specimens of clinical female breast cancer patients and their clinicopathological data were collected.Normal breast epithelial cells MCF-10 A and breast cancer cell lines MCF-7,MDA-MB-231,SK-BR-3 were cultured.Real-time quantitative PCR was used to detect the expression of GPR120 mRNA in breast cancer tissues,adjacent normal tissues and breast cancer cell lines and normal breast epithelial cells.Analyzing the relationship between the expression of GPR120 mRNA in breast cancer tissues and the clinicopathological characteristics of breast cancer patients.2.Western blot was used to detect the expression of GPR120 protein in normal breast epithelial cells MCF-10 A and breast cancer cell lines MCF-7,MDA-MB-231,SK-BR-3.Using AH7614 the selective inhibitor for GPR120 to treat MCF-7,Plate clone formation assay,CCK8 assay,Wound Healing assay,and Transwell assay were used to observe the proliferation,migration and invasive ability of MCF-7 after GPR120 inhibited.Results : 1.The mRNA expression level of GPR120 in breast cancer tissues was significantly lower than that in adjacent tissues(P<0.05).Expression of GPR120 mRNA in breast cancer cells MCF-7,MDA-MB-231 and SK-BR-3 were significantly lower than that in normal breast epithelial cells MCF-10A(P<0.05).2.GPR120 mRNA expression in the EGFR negative group was significantly higher than that in the EGFR positive group(P<0.05).Compared to general invasive carcinoma and special invasive carcinoma,the mRNA expression of GPR120 in non-invasive carcinoma was significantly higher than that in the two groups(P<0.05).There was no significant difference in GPR120 gene expression between general invasive carcinoma and special invasive carcinoma(P>0.05).3.GPR120 protein expression of breast cancer cells MCF-7 was significantly higher than that of normal breast epithelial cells MCF-10 A.When the concentration of AH7614 was 7?M,the expression of GPR120 protein in MCF-7 cells was significantly inhibited.The results of Plate clone formation assay and CCK8 assay showed that compared with the untreated group,the cloning formation rate of MCF-7 in the treated group decreased significantly(P<0.05).The results of Wound Healing assay and Transwell experiment showed that the migration distance and across cell membranes of MCF-7 cells in the AH7614 group were significantly lower than those in the untreated group(P< 0.05).Conclusion:The expression of GPR120 mRNA in breast cancer tissues was lower than that in adjacent tissues,and its expression was correlated with the expression of EGFR and the pathological classification of breast cancer.The expression of GPR120 protein in breast cancer cells was higher than that in normal breast epithelial cells.The gene expression of GPR120 was inconsistent with protein expression.GPR120 protein promotes the proliferation,migration and invasion of breast cancer cells MCF-7.
