Objective By detecting EBV infection and TCAB1 protein expression in laryngeal Squamous Cell Carcinomas tissues,the correlation between TCAB1 protein expression and laryngeal Squamous Cell Carcinomas was analyzed,and the mechanism of TCAB1 protein overexpression in the occurrence and development of EBV-positive laryngeal cancer was preliminarily explored,which provided a theoretical basis for early diagnosis and early treatment of laryngeal cancer.Methods EBV gene detection and immunohistochemical qualitative analysis of TCAB1 protein were carried out in 113 paraffin tiss?e specimens of laryngeal Squamous Cell Carcinomas from the Department of Otolaryngology,Affiliated General Hospital of Ningxia Medical university from 2011 to 2017 and 12 fresh specimens of laryngeal Squamous Cell Carcinomas collected from Pujiang Branch of Shanghai Wuguan Hospital in 2018.Eight eligible specimens were selected for quantitative analysis of Western-blotting TCAB1 protein.The survival of patients with laryngeal squamous cell carcinomas after operation was analyzed.Results(1)The majority of laryngeal squamous cell carcinomas patients were male(122/125,97.6%).Most of them were aged from middle-aged to young and elderly(45-74 years old,86.4%).Most of them were located in glottic region(92/125,73.6%).The course of disease was within one year(91/125,72.8%);(2)The positive rate of EBV infection in laryngeal cancer was 61.6%(77/125),and had no significant correlation with age,sex,nationality,course of disease,differentiation degree of laryngeal cancer tissue,clinical stageand location of tumor(P > 0.05);(3)TCAB1 protein was mainly expressed in the nucleus of laryngeal cancer tissues,followed by a small amount in the cytoplasm;(4)The expression of TCAB1 protein in laryngeal cancer tissues was associated with EBV infection(P < 0.05).The expression of TCAB1 protein in EBV-positive laryngeal cancer tissues,EBV-negative laryngeal cancer tissues,EBV-positive adjacent tissues and EBV-negative adjacent tissues showed a gradual decline trend;(5)The expression of TCAB1 protein in laryngeal squamous cell carcinomas tissues was associated with EBV infection(P < 0.05).The expression of TCAB1 protein in EBV-positive laryngeal squamous cell carcinomas tissues,EBV-negative laryngeal squamous cell carcinomas tissues,EBV-positive adjacent tissues and EBV-negative adjacent tissues showed a gradual decline trend;(6)Epstein-Barr virus infection and TCAB1 protein expression were not significantly correlated with the prognosis of laryngeal cancer patients after operation(P > 0.05),but the location of tumors was an independent risk factor for the prognosis of laryngeal squamous cell carcinomaspatients after operation.Conclusion This study found that EBV infection and TCAB1 protein expression were closely related to the occurrence and development of laryngeal squamous cell carcinomas,and the expression of TCAB1 protein in EBV-positive laryngeal squamous cell carcinomas,EBV-negative laryngeal squamous cell carcinomas,EBV-positive adjacent tissues and EBV-negative adjacent tissues showed a gradual decline trend.It is suggested that TCAB1 gene is involved in the pathogenesis of EBV-positive laryngeal cancer.This provides a new idea and perspective for targeted therapy of EBV-positive laryngeal squamous cell carcinomas.
