Objective: To investigate the effects of MEX3 C gene and insulin-like growth factor-1(IGF-1)on follicular development and proliferation in ovarian granulosa cells of the mice,and to clarify the relationship between MEX3 C gene and IGF-1 in ovarian granulosa cells.To elucidate the mechanism of MEX3 C gene mediating IGF-1 on proliferation of the ovarian granulosa cells in mice.Methods: Thirty female C57 mice were randomly divided into two groups: control group and IGF-1 group.The morphological changes of follicles were observed by HE staining and the expression of insulin-like growth factor-1 receptor(IGF-1R)was detected by immunohistochemistry.The ovarian granulosa cells in mive were cultured and amplified in vitro and transfected with lentivirus MEX3 C sh RNA.Then the granulosa cells before and after transfection were cultured by IGF-1 in vitro.The cells were divided into six groups:blank control group,MEX3 C sh RNA control group,MEX3 Csh RNA group,I GF-1 culture group,MEX3 Csh RNA control combined with IGF-1 group,MEX3 Csh RNA combined with IGF-1 group.The expression of MEX3 C protein in normal mouse ovarian granulosa cells was verified by immunofluorescence.Western Blot was used to detect the expression of MEX3 C protein and proliferating cell nuclear antigen(PCNA),IGF-1R,protein kinase B(AKT),phosphorylated protein kinase B(P-AKT).SPSS21.0 statistical software analysis,P<0.05 means the difference was statistically significant.Results: 1 MEX3 C protein was expressed in cytoplasm and nucleus of the ovarian granulosa cells in mice.2 Ovarian granulosa cells showed green fluorescence in MEX3 C sh RNA control group and MEX3 C sh RNA group.3 IGF-1 group showed significantly larger follicles.The expression of IGF-1R protein in IGF-1 culture group was significantly higher than that in control group(P<0.05),and the expression of PCNA protein in culture group was significantly higher than that in blank control group(P<0.05).5 The expression of MEX3 C protein and PCNA,IGF-1R,P-AKT protein in MEX3 C sh RNA group were significantly lower than those in blank control group and MEX3 C sh RNA control group(all P<0.05).The expression of IGF-1R,PCNA,P-AKT protein in MEX3 C sh RNA + IGF-1 group and MEX3 C sh RNA + IGF-1 group was significantly higher than that in blank control group,MEX3 C sh RNA control group and MEX3 C sh RNA group(all P<0.05).Conclusions: 1 MEX3 C protein was localized in the cytoplasm and nucleus of ovarian granulosa cells.2 IGF-1 promoted follicular development and granulosa cell proliferation in mice.3 MEX3 C gene enhanced the proliferation of granulosa cells.4 MEX3 C gene regulated IGF-1R and stimulated the proliferation of granulosa cells through phosphorylaed AKT.
