Objective Through the selfmade cervical anteflexion device,rabbit skeletal muscle static property damage,watch back to reinforcement technique for the damage to the sk-eletal muscle peripheral pain medicine material,histomorphology,muscle of cytoskeleton proteins Desmin and a-Actin expression,the effect of back from organization and cell biology to explore medical reinforcement technique in treatment of static possible mechanisms of skeletal muscle injury.Methods Sixty rabbits randomly selected 18 for the Normal group(Normal,Nor group),the remaining 42 after successful skeletal muscle static damage Model is establi-shed,according to random number table method is divided into Model group(Model s-et,MS group,6),back to the medicine with muscle group(Li jin,LJ group,18)and nat-ural recovery group(Self repairing,the SR group,18).At the end of modeling,li jin g-roup received manipulation treatment,while the normal group and the natural recovery group did not receive treatment.Six rabbits were randomly selected at 7 days,14 days and 21 days after injury,respectively.Blood was collected from the ear vein,and the contents of 5-HT,?-EP and SP were determined by enzyme-linked immunosorbent assay.The ultrastructure of tissue was observed by transmission electron microscopy,histopath-ological changes were observed by HE staining,positive muscle fibers were measured by immunohistochemistry,and the expression levels of Desmin and ?-Actin were detect-ed by western blot.SPSS22.0 statistical software was used to analyze the experimental data.Results1.Model evaluationChanges in the degree of cervical muscle stiffness:during the modeling process,the neck muscle tissue of the model group gradually became stiff,and nodules or cords co-uld be touched after the modeling,indicating that the rabbit model of skeletal muscle static injury was successful.Muscle histomorphological changes:from the longitudinal view,most of the muscle fibers were disorganized,and the muscle fibers were distorted and fractured,with a large number of inflammatory cells infiltrating,and muscle cells atrophic and muscle cell s-pace significantly widened on the transverse view,indicating that the rabbit model of s-keletal muscle static injury was successful.2.General conditions:The rabbits in each group survived until the end of the expe-riment.Before the end of modeling,the experimental rabbits were sensitive,had vivid ey-es,and had obvious escape reaction to capture.After modeling,the mental state is gradu-ally not good,hair loose and dull luster,thin body,reduced diet.3.Peripheral blood test indicators:?-EP test results:After modeling,there was no sta-tistically significant change in ?-EP content in the model group,the li jin group and t-he natural recovery group(P>0.05),indicating that the experiment was comparable.Co mpared with the normal group,the ?-EP content was significantly reduced(P<0.05).The-re was a statistically significant difference in ?-EP content between the two groups before and after treatment(P<0.05).After injury treatment for 14 days,the ?-EP contentin the li jin group was higher than that in the natural recovery group(P<0.05).After injury treatment for 21 days,the ?-EP content in the li jin group was significantly high-er than that in the natural recovery group(P<0.05).5-HT test results:In the model group,the li jin group and the natural recovery gro-up,there was no statistically significant change in 5-HT content after modeling(P>0.05),indicating that the experiment was comparable.Compared with the normal group,the5-HT content was significantly increased(P<0.05).There was a statistical difference in5-HT content between the two groups before and after treatment(P<0.05).After injury treatment for 7 days,the 5-HT content in the li jin group was lower than that in the natural recovery group(P<0.05).After injury treatment for 14 days,the content of 5-HT in the li jin group was significantly lower than that in the natural recovery group(P<0.05).After injury treatment for 21 d,there was no significant difference in 5-HT content between the li jin group and the natural recovery group(P>0.05).SP test results:After modeling,there was no significant change in SP content in th-e model group,the li jin group and the natural recovery group(P>0.05),indicatin that the experiment was comparable.Compared with the normal group,SP content was signif-icantly increased(P<0.05).There was a statistical difference in SP content between the two groups before and after treatment(P<0.05).After injury treatment for 7 days,the S-P content in the li jin group was lower than that in the natural recovery group(P<0.05).After injury treatment for 14 days,the SP content in the li jin groupwas significant ly lower than that in the natural recovery group(P<0.05).After injury treatment for 21 days,there was a significant difference in SP content between the li jin group and the natural recovery group(P<0.05).4.Histopathological observation:Results of HE staining:muscle cells in the normal group were orderly arranged,with regular shape and complete structure.In the model group,most of the muscle fibers were disordered,and the muscle fibers were twisted,defo-rmed and fractured,with the transverse striae disappeared,a large number of inflammat ory cells infiltrated,muscle cells atrophy,and the muscle cell space was significantly widened.After injury treatment for 7 days,the muscle fibers in the treatment group and t he natural recovery group were obviously broken and twisted,with inflammatory cell in filtration and a small number of vacuolar myocytes.After injury treatment for 14 days,muscle fiber swelling and inflammatory cell infiltration still existed in the treatment gr-oup of li jin manipulation,but the muscle fiber structure was significantly improved compared with the natural recovery group.After injury treatment for 21 days,the muscle cells in the li jin manipulation group were orderly and tight,with new muscle cells,and the swelling of muscle fibers significantly subsided.In the natural recovery group,there was still a small amount of inflammatory cells infiltrating,and the muscle fiber structur-e improved significantly.Ultrastructure of muscle fibers was observed by transmission electron microscopy:A large number of muscle fibers composed of dense and neatly arranged myofilaments were observed in group normal and complete z-line and m-line could be clearly seen.I-n themodel group,the muscle fibers were loose,twisted and deformed and fractured,and the intact sarcomere could not be found.The M line disappeared and the Z line partial-ly disappeared.After 7 days of injury treatment,the muscle fibers of the li jin manipul-ation group were loose,and a small part of the intact myoculi could be seen,and part of the z-line could be clearly seen.The width and size of the myoculi of the natural r-ecovery group were different,and the muscle fibers were obviously loose,and parof the z-line could be seen.After injury treatment for 14 days,the muscle fibers ine li jin manipulation group were loose,most of which were clear z-line and mline.Alarge number of mitochondria were evenly distributed among the muscle fibers,and estructure was in tact.The degree of muscle fiber loose in the natural recovery group washeavier than th-at in the li jin manipulation group.Injury treatment of 21 days,back to the medicine with reinforcement technique most complete set of sarcome,muscle fi-ber density,a sma-ll number of lines to Z and M still have damage,muscle fiber improved markedly,natural recovery group has a small part of the muscle fibers in distorted state,Z line is c-lear,a small amount of sarcomere,complete structure of a large number of mitochondri-a gathered in muscle fiber damage.5.Skeletal muscle tissue Desmin and a-Actin positive muscle fiber(AODvalue):co-mpared with the model group,the normal group had higher Desmin and a-Actin muscl-e fiber AOD value(P<0.05).The AOD values of Desmin and a-Actin muscle fibers in the li jin group increased gradually at 7 days and 14 days,14 d ays and 21 days(P<0.05),while those in the natural recovery group increased at 14 days and 21days(P<0.05).The AOD values of Desmin and a-Actin positive muscle fiber in the li jin group were higher than those in the natural recovery group(P<0.05)and those in the li ji-n group were higher than those in the natural recovery group(P<0.05)and those in the li jin group were higher than those in the natural recovery group(P<0.05).6.Expression of Desmin and A-actin in skeletal muscle tissues was significantly h-igher in the normal group than in the model group(P<0.05).The expression levels of Desmin and A-actin were increased gradually between each group at 7 days and 14 d-ays,14 days and 21 days in the li jin group(P<0.05),while the expression levels of Desmin and A-actin were increased between the natural recovery group at 14 days and 21 days(P<0.05).7 days injury treatment,back muscle group of skeletal muscle fiber Desmin,A-actin medicine with Actin expression level is higher than natural recovery gr-oup(P<0.05),14 days injury treatment,back muscle group of skeletal muscle fiber Des-min,A-actin medicine with Actin expression level is higher than natural recovery grou-p(P<0.05),21 days injury treatment,back muscle group of skeletal muscle fiber Desmi-n,A-actin medicine with Desmin expression level is higher than natural recovery group(P<0.05).Conclusion1.By adjusting the contents of ?-EP,5-HT and SP in peripheral blood,li jin manip-ulation achieves analgesic effect.2.By increasing the expression of cytoskeleton protein Desmin and alpha-actin,li jin can promote the repair of injured skeletal muscle tissue morphology and structure.
