Objective:In this study,rats with Chronic Constriction Injury(CCI)were used to induce ne uropathic pain,and the gastrocnemius of diseased side was treated with traditional Chi neseTuina.The Paw withdrawal threshold,Paw withdrawal latency,CatWalk Three-dim ensional gait,HE pathological staining and Western blotting were used to observe the thenalgesic effect of massage on CCI model rats and the changes of SP/NK-1R in ra t dorsal root ganglia.The SP/NK-1R pathway involved in the analgesic effect of the massage point method on CCI model rats.Method:1 32 120-180 g male Sprague-Dawley rats were randomly divided into 4 groups(8 in each group),weighed and numbered,respectively,naive group,sham operation g roup,CCI model group and CCI+Tuina group.Rats in the CCI+Tuina group were inv olved in the Tuina method on the 4th day after modeled,10 minutes/time/day,and co ntinuous intervention for 14 days.The changes of Paw withdrawal threshold,Paw wit hdrawal latency were observed before and during 1,3,7,10,14 and 17 days after m odeled.After collecting the behavioral indexes of each group of rats,the rats were kill ed andthe right L4 and L5 dorsal root ganglia were removed.HE pathological stainin g was used to observe the right dorsal root ganglia of each group 17 days after mode l establishment and to observe changes in morphological structure.2 32 120-180 g male Sprague-Dawley rats were randomly divided into 4 groups(8 in each group),weighed and numbered,respectively,naive group,sham operation g roup,CCI model group and CCI+Tuina group.Rats in the CCI+Tuina group were inv olved in the Tuina method on the 4th day after modeled,10 minutes/time/day,and co ntinuous intervention for 14 days.The changes of the three-dimensional gait of CatWa lk before,during 1,3,7,10,14 and 17 days after modeled were observed.After coll ectingthe behavioral indexes of each group of rats,the rats were killed and the rightL4 andL5 dorsal root ganglia were removed.Western blotting was performed.The right dorsal root ganglion SP and NK-1R content of each group was observed 17 days after model establishment.Result:1 Paw withdrawal threshold,Paw withdrawal latency: CCI modeling can significa ntlyreduce PWL and PWT values in rats,which is significant compared with the naiv e group(P<0.001).The PWL value of CCI+Tuina group was statistically significant on the 10 th day after modeled(P<0.05);the PWT value of CCI+Tuina group was st atistically compared with the CCI model group on the 10 th,14th and 17 th day after modeled.Academic significance(P<0.05).2 Catwalk gait parameters: Maxcontact area,print area,swing speed,and stand of CCI model group and the CCI+Tuina group decreased significantly after CCI modele d,and the swing increased,which was significantly different from the naive group(P<0.001).The print area,maxcontact area and swing CCI+Tuina group were statisticall y different from CCI model group in the 14 th and 17 th day after modeled(P<0.05,P<0.01);swing speed,the differences were significant on CCI+Tuina group and CCI model group in the 3rd,14 th and 17 th day after CCI modeled(P<0.05,P<0.01).The stand,CCI+Tuina group was statistically significant compared with CCI model group i n the 3rd and 10 th day after modeled(P<0.05).3 HE staining of dorsal root ganglion: The dorsal root ganglia of CCI+Tuina gro up was similar in structure to the CCI model group.Compared with the naive group and the sham operation group,the phenomenon of neuronal dissolution,rupture and ne crosis was found,there was no significant difference between these two groups.4 SP/NK-1R expression: The content of SP/NK-1R protein in the right L4 and L5 DRG neurons of the CCI model group was significantly different from that of the na ive group,the sham operation group,and the CCI+Tuina group(P<0.001,P<0.001,and P=0.006<0.05).Conclusion:1 Tuina can reduce the mechanical Paw withdrawal threshold,Paw withdrawal lat encyon CCI rats,improve the three-dimensional gait of CatWalk on rats,and have ag ood analgesic effect.But for rat dorsal root,there was no major change in the morph ology of ganglion cells.2 The dorsal root ganglion SP/NK-1R pathway may be involved in the analgesic process of Tuina for the treatment of neuropathic pain.
