Objective:To investigate the expression and clinical significance of KLF12 in breast tumors.Method:Breast cancer tissues surgically removed and pathologically confirmed from the pathology department of Shaanxi provincial tumor hospital from January 2014 to July 2017 were selected.All cases must have complete case data and are the first time detection of breast cancer patients,the original specimens were large specimens removed surgically.All of the patients has not been preoperative radiation and chemotherapy and endocrine therapy.According to the expression of ER,PR and HER2 in the pathological report of breast cancer patients(If ER,PR and HER2 were not detected,we will carry out in this experiment),100 triple-negative breast cancer(TNBC)patients and 100 non-triple-negative breast cancer(NTNBC)patients were randomly selected,and normal tissues adjacent to cancer were selected as controls.Immunohistochemistry(IHC)was used to detect the expression of KLF12 in 200 cases of breast cancer.The relationship between the expression of KLF12 and the clinicopathological characteristics of breast cancer was analyzed by statistics.Results:1.Among the 200 cases of breast cancer,159 cases had KLF12 positive expression,with a positive rate of 79.5%(159/200)and 41 cases had negative expression.Among the200 normal tissues,only 18 cases had KLF12 positive expression,and the positive rate was only 9%(18/200).The remaining 182 cases were all negative.The results showed that the expression of KLF12 in breast cancer was significantly higher than that in control group,and the expression of KLF12 in breast cancer tissues and control group was different,with statistical significance(P < 0.05).2.Among the 100 cases of TNBC,there were 91 cases with KLF12 positive expression,and the positive rate was 91%(91/100).Among them,there were 9 cases with a score of 0,6 cases with a score of 1,33 cases with a score of 2,and 52 cases with a score of3.The proportion of each score is: 0 is 9%(9/100),1 is 6%(6/100),2 is 33%(33/100),and 3is 52%(52/100).Among the 100 cases of NTNBC,there were 68 cases with positive KLF12 expression,and the positive rate was 68%(68/100),among which 32 cases had a positive score of 0,25 cases had a score of 1,31 cases had a score of 2,and 12 cases had a score of 3.The proportion of each score is: 0 is 32%(32/100),1 is 25%(25/100),2 is 31%(31/100),and3 is 12%(12/100).The results showed that the positive proportion of KLF12 in the breast cancer of the TNBC group was higher than that of the NTNBC group.Statistical analysis showed that KLF12 expression in the TNBC group was different from that of the NTNBC group,which was statistically significant.(? 2 = 49.610,P = 0.000).3.To further analyze the correlation between the expression of KLF12 and the age,size,histological type,TNM stage,lymph node metastasis and ER?PR? HER2?Ki67expression of breast cancer.The results showed that KLF12 expression was correlated with lymph node metastasis of breast cancer?HER2 expression?Ki67 expression.(P< 0.05).There was no correlation with age,size,TNM stage and histological grade of breast cancer(P >0.05).Conclusion:1.The high expression of KLF12 in breast cancer suggests that KLF12 may be helpful in the diagnosis of breast cancer.2.The high expression of KLF12 in TNBC suggests that breast cancer with high expression of KLF12 may be more malignant and aggressive.3.The expression of KLF12 in breast cancer has no correlation with age,size,pathological type,TNM stage,histological grade of breast cancer and expression of ER?PR,but has statistical significance with lymph node metastasis?HER2 and Ki67,suggesting that the high expression of KLF12 may promotes the metastasis and poor prognosis of breast cancer.
