Objective:By detecting the changes of the expression levels of RNA-binding motif protein 5?RBM5?and Beclin1 in clear cell renal cell carcinoma?ccRCC?tissue,para-carcinoma tissue and normal renal tissue,the objective of this study was to preliminarily explore the clinical significances of RBM5 and Beclin1 in ccRCC based on the clinical data.Methods:Following surgical resection,67 specimens of ccRCC tissues confirmed by pathological diagnosis,25 specimens of para-carcinoma tissue and 10 specimens of normal renal tissue were paraffin-embedded before preparing tissue slices.The pathological change of each tissue was observed by hematoxylin-eosin staining?HE?,and the changes of RBM5 and Beclin1 protein expression in each tissue were detected by immunohistochemistry?IHC?to analyze their relationships with relevant clinical parameters.Results:1.Pathological changes of ccRCC tissue,para-carcinoma tissue and normal renal tissue?HE staining??1?ccRCC tissue:Microscopic observation suggested that the cytoplasm of the tumor cells were transparent and tumor cells were arranged in nests.The reticular interval can be seen and there were tumor vessels and blood sinuses between them;tumor cells were large in different forms;the cytoplasm staining of tumor cells was relatively shallow,and nuclear morphological variation and pyknosis were noted;tumor cell heteromorphism became more evident with the clinical stage.?2?Para-carcinoma tissue:Microscopically,the renal tissue morphology was basically normal,the glomeruli and renal tubules were normal in structure;cells were distributed evenly,cytoplasm was stained stronger,and the tissue was not involved by tumor.?3?Normal renal tissue:Microscopically,the renal tissue morphology was normal,and the glomeruli and renal tubules were normal in structure.2.Changes of RBM5 and Beclin1 expression in ccRCC tissue,para-carcinoma tissue and normal renal tissue2.1 RMB5 protein?1?RMB5 protein was expressed mainly in nuclei and cytoplasm.The expression level of RBM5 protein in ccRCC tissue was apparently lower than those in para-carcinoma tissue and normal renal tissue?P<0.05?.The expression level of RBM5 protein in para-carcinoma tissue was significantly higher than that in normal renal tissue?P<0.05?.?2?In ccRCC tissue,the expression level of RBM5 was significantly associated with the clinical stage of the tumor,lymphatic metastasis,distant metastasis,pathological grading and tumor size?P<0.05?,but had no obvious correlation with sex,age,bodyweight,smoking history and history of hypertension?P>0.05?.In para-carcinoma tissue,the expression level of RBM5 was significantly associated with the clinical stage of the tumor,lymphatic metastasis and tumor size?P<0.05?,but had no obvious correlation with sex,age,bodyweight,smoking history,history of hypertension,distant metastasis and pathological grading?P>0.05?.2.2 Beclin1 Protein?1?Beclin1 protein was primarily expressed in cytoplasm.The expression levels of Beclin1 in para-carcinoma tissue and normal renal tissue were remarkably higher than that in ccRCC tissue?P<0.05?.The expression level of Beclin1 in para-carcinoma tissue was much higher than that in normal renal tissue?P<0.05?.?2?The expression level of Beclin1 in ccRCC tissue was significantly correlated with the clinical stage of the tumor,lymphatic metastasis and tumor size?P<0.05?,but had no obvious correlation with sex,age,bodyweight,smoking history,history of hypertension,distant metastasis and pathological grading?P>0.05?.The expression level of Beclin1 in para-carcinoma tissue was significantly associated with the clinical stage of the tumor,lymphatic metastasis and tumor size?P<0.05?,but had no obvious correlation with sex,age,bodyweight,smoking history,history of hypertension,distant metastasis and pathological grading?P>0.05?.3.Correlation between RBM5 and Beclin1The correlation between RBM5 expression and Beclin1 expression in ccRCC tissue and para-carcinoma tissue were explored by a Pearson's correlation analysis.The result revealed that RBM5 expression was positively correlated with Beclin1 expression,with statistical significance?r1=0.57,r2=0.92,P<0.05?.Conclusions:1.The expression levels of both RBM5 and Beclin1 are down-regulated in ccRCC tissue and up-regulated in para-carcinoma tissue,obviously higher than those in normal renal tissue and tumor tissue,.2.There is a significant correlation between RBM5 and Beclin1 and ccRCC clinical stage,lymph node metastasis and tumor size.3.In ccRCC tissues and adjacent tissues,RBM5 is positively correlated with the expression of Beclin1.RBM5 and autophagy may have synergistic effects in the process of affecting ccRCC.combined detection of both expression in ccRCC tissue is expected to be a new indicator for assessing ccRCC progression and a potential therapeutic target for controlling ccRCC progression point.
