Objective:To study the effect of erythropoietin on cardiac macrophages in allograft ratsMethods:18 pairs of SD rats with successful allogeneic heart transplantation(HT)were successfully performed.According to different treatment methods,they were divided into three groups:group A(transplantation group),only heart transplantation;group B(treatment group),treatment with erythropoietin(EPO)after heart transplantation;group C(inhibitor group),after the heart transplantation,treatment with macrophage inhibitor chlorophosphate liposome.Rats in each group were sacrificed 2 days after heart transplantation,and the donor apex tissue was taken,and blood samples were taken from the apex of the donor heart.Heart tissue was stained with HE to observe the morphological structure of myocardial tissue;Elisa kit was used to detect troponin I(Tn I)in the serum of recipient rats;immunohistochemistry was used to detect macrophages in myocardial tissue.The degree of expression quantitative analysis of macrophage content in myocardial tissue by Western Blot(WB).Results:1.The results of HE staining in each group were compared:the myocardial structure of group A was loose,the cell spacing was widened,and the number of nuclear blue staining was more.The myocardial structure of group B was relatively intact,the cell spacing was neat,the nucleus blue stain was less,and the cytoplasm was red stained.Myocardial cells in group C are disordered in structure,large in cell gap,and large in number of nuclear blue stains.2.Comparison of the results of troponin I in blood of each group:ELISA detection of serum troponin I in rats showed that the measured value of troponin I in group A was0.135±0.005 higher than that in group B 0.094±0.007,statistically significant(~#p<0.05);group B troponin I measured value of 0.094±0.007 lower than C group 0.165±0.008,the difference was statistically significant(~*p<0.05);group C measured value of 0.165±0.008 higher than Group A was 0.1350.005,the difference was statistically significant(~?p<0.05).3.Comparison of immunohistochemical detection of semi-quantitative IOD values of F4/80 macrophages:semi-quantitative IOD values in group A were 25.29±1.86 higher than those in group B 18.36±1.12,the difference was statistically significant(~#P<0.05);group B semi-quantitative IOD values was 18.36±1.12 higher than that of C group 10.21±1.23,the difference was statistically significant(~*P<0.05);the semi-quantitative IOD value of group C was 10.21±1.23 lower than that of group A 25.29±1.86,the difference was Statistical significance(~?P<0.05).4.Comparison of quantitative results of F4/80 macrophages detected by Western blot:The results were detected by Western blotting of the macrophage marker F4/80 to reflect the number of macrophages.The quantitative value of group A was 0.450±0.041 higher than that of group B by 0.351±0.033,the difference was statistically significant(~#P<0.05);the quantitative value of group B was 0.351±0.033 higher than that of group C by 0.259±0.036,the difference was statistically significant(~*P<0.05);the quantitative value of group C was lower than that of group A by 0.450±0.041,the difference was statistically significant(~?P<0.05).Conclusion:Effect of EPO on cardiac macrophage infiltration in acute allograft rats...
