Objective: To investigate the effects of renal ischemia pre-irrigation on hepatic inflammatory factors and oxidative stress.Methods: Eighteen healthy male Sprague-Dawley rats were randomly divided into three groups: ischemia-reperfusion injury non-irrigation group(group A),ischemia-reperfusion injury pre-irrigation group(group B),and sham operation group(group C)(6 rats/ group).The right kidney was removed from group A and group B,and the left renal pedicle was clipped with non-invasive microvascular clips for 60 minutes and then released for blood reperfusion.Group B was given rapid saline lavage before ischemia to reduce residual blood cells in renal blood vessels;group C only removed the right kidney in the open stomach,and the left kidney was not treated with ischemia.The rats were sacrificed 24 hours after reperfusion and the inferior vena cava blood,left kidney and liver specimens were collected for study:1.Automatic biochemical analyzer detects serum creatinine,blood urea,alanine aminotransferase(ALT)and aspartate aminotransferase(AST);2.HE staining: The pathological changes of kidney and liver tissues of rats in each group were observed.3.Immunohistochemistry was used to detect heme oxygenase-1(HO)in the liver tissue of each group,monocyte chemoattractant-1(MCP-1)and interleukin-6(IL-6)expression;4,real-time PCR detection of catalase in the liver tissue of each group of rats(CAT),superoxide dismutase(SOD),tumor necrosis factor alpha(TNF-?),IL-6 and MCP-1 mRNA expression.Results:(1)18 models successfully survived and survived,and 2 failed in modeling failure,2 were made,and the mortality rate was 10%.(2)Renal function: serum creatinine and urea were significantly higher in group A and group B than in group C(P<0.05),andgroup B was lower than group A(P<0.05).(3)Liver function: Serum ALT and AST in group A were significantly higher than those in group B and C(P<0.05),and the expression of group C was lower than that in group B(P<0.05).(4)HE staining of renal tissue: In group A,the renal capsule was dilated,the renal glomerulus was atrophied,the tubular lumen was dilated,the epithelial cells of the proximal convoluted tubule were enlarged,the cytoplasm was loose,and the renal interstitial edema.In group B,the renal glomeruli were slightly atrophied,the volume was slightly smaller,the renal capsule was enlarged,and some of the tubular epithelial cells were enlarged.There was no abnormality in the renal structure in group C.(5)HE staining of liver tissue: In the liver tissue of group A,hepatic sinusoids were obviously dilated and congested,hepatic cell cords were disordered,hepatocyte cytoplasm staining became shallow and vacuoles appeared,hepatocyte edema was obvious,volume increased,and nuclear staining became shallow.In the liver tissue of group B,hepatic cell cords were arranged negligibly and the hepatic sinusoids were slightly expanded.In the liver tissues of group C,the hepatic cells are arranged neatly and uniformly,and the hepatocytes have no obvious swelling and edema.(6)Compared with group C,the expression of MCP-1 and IL-6 in liver tissues of group A and group B was significantly increased(P <0.05),and the expression of group B was lower than that of group A(P <0.05).The highest expression in group B was higher than that in group A and C(P <0.05),while the expression in group A was higher than that in group C(P<0.05).(7)The mRNA levels of MCP-1,TNF-? and IL-6 were the least in group C,and the expressions in group A and B were significantly higher(P<0.05),while group A was higher than group B(P).<0.05);SOD and CAT mRNA levels were significantly up-regulated in group B,compared with group A and C(P<0.05);group A was significantly lower than group C(P<0.05).Conclusion: 1.Rat kidney ischemic pre-irrigation can reduce the production of inflammatory factors in the liver and protect the liver;2.Rat kidney ischemic pre-irrigation can increase the content of antioxidants in theliver,weaken the oxidative stress response and protect the liver.
