Objective:To study the inhibitory effect of aspirin on the growth of gastric cancer cells through induction of autophagy,and preliminarily analyze its possible mechanism of action.Methods:?1?Human gastric cancer cells SGC-7901,BGC-823 were treated with different concentrations of aspirin,cell viability was detected by MTT assay;?2?The gastric cancer cell Apoptosis and cell cycle were detected by flow cytometry;?3?Nuclear morphological changes were detected by Hoechst33258 staining;?4?Transwell migration test was used to detect cell migration;?5?Confocal laser scanning microscopy was used to detect the formation of autophagosomes;?6?Western blot was used to detect the expression of LC3-?protein.Results:?1?MTT assay show that aspirin had a significant inhibitory effect on gastric cancer cells SGC-7901 and BGC-823,and the cell viability rate decreased with the increase of aspirin drug concentration?1mM,2mM,4mM,6mM,8mM,10mM?(IC50was about 6mM,P<0.01).?2?The results of flow cytometry showed that aspirin could not be treated within 48h induced cell apoptosis but induced cell cycle arrest in G1phase?P<0.05?;?3?The results of Hoechst33258 staining showed that the DNA in gastric cancer cell nucleus was not fragmented by enzymatic cleavage,no apoptotic bodies were observed,and no luminescent bright blue color was observed,consistent with flow cytometry results of apoptosis.?4?Transwell migration test show that SGC-7901 CK group 201.67±6.51?5mM group 96.67±6.43,BGC-823 CK group96.00±4.58?5mM group 45.67±7.09.aspirin can significantly inhibit the migration of two gastric cancer cell lines?P<0.001?.?5?Confocal laser scanning microscopy and Western blot showed that after treatment with aspirin,the number of autophagosomes and the expression of autophagy protein LC3-?were increased,and the autophagy is activated.Conclusion:?1?Aspirin significantly inhibited the proliferation of human gastric cancer cell SGC-7901 and BGC-823 with a significant dose-response relationship;?2?Aspirin can arrest the cell cycle of human gastric cancer cells SGC-7901 and BGC-823 in G1 phase,and its mechanism of action may be related to the activation of cell autophagy.
