Objective:In recent years,studies have found that oncogene-induced cell senescence is an important event in tumorigenesis.This study aimed to investigate the activation of K-ras in gastric cancer and precancerous lesions,the relationship between K-ras and aging indicators,and K-ras.The difference of induced cell senescence in gastric cancer and precancerous lesions provides a theoretical basis for clinical therapy of gastric cancer.Methods:A total of 138 specimens of paraffin-embedded tissue of gastric tubular adenocarcinoma with complete surgical records from 2016 to 2017 in the Department of Pathology,Yan'an University Hospital of Shanxi Province were collected.All specimens were treated with pathologists of two deputy directors and above according to the new WHO gastric cancer.Classification criteria for diagnosis.All the selected cases were found to have gastric cancer for the first time.No radiochemotherapy or immunotherapy was performed before the"radical gastrectomy",all of the paracancerous tissues showed precancerous lesions such as chronic atrophic gastritis with intestinal metaplasia or intraepithelial neoplasia.The cancer tissues and adjacent tissues of gastric cancer patients?276 wax blocks?were used as the research object.EnVision immunohistochemical staining was used to detect the expression of K-ras,P16INK4a,P53 and SA-?-Gal in all tissues.The situation was analyzed.The difference of K-ras expression between cancer tissues and paracancerous tissues was analyzed.The relationship between K-ras and P16INK4a,P53 and SA-?-Gal senescence indicators was discussed in cancer tissues and adjacent tissues and compare the differences of OIS in different organizations.Results:1.The positive rate of K-ras expression in cancer tissues was 95.7%,and the positive rate of K-ras expression in adjacent tissues was 3.6%.The expression of K-ras in cancer tissues was significantly higher than that in adjacent tissues.The difference was statistically significant?P<0.05?.2.There are 138 wax blocks in the cancer tissue,including 132 in the k-ras positive group and 6 in the negative group.The positive rates of P16INK4a,P53 and SA-?-Gal in K-ras positive group?66.7%,77.3%,87.9%?were higher than those in K-ras negative group?16.7%,33.3%,33.3%?.The difference was statistically significant?P<0.05?.The K-ras gene in cancer cells is positively correlated with aging indicators,indicating that K-ras can induce cellular senescence in cancer cells.3.All the 138 wax blocks of adjacent tissues were divided into k-ras positive group and negative group,including 5 positive group and 133 negative group.The positive rates of P16INK4a,P53 and SA-?-Gal in K-ras positive group?80.0%,60.0%,80.0%?were higher than those in K-ras negative group?4.5%,2.3%,3.8%?.The difference was statistically significant?P<0.05?.Precancerous lesions of gastric cancer were found in all the selected adjacent tissues,K-ras gene was positively correlated with aging index in precancerous lesions,indicating that K-ras induced cell senescence in precancerous lesions.4.Cell senescence induced by K-ras in cancer tissues was significantly higher than that in adjacent tissues.Conclusion:1.There is a difference in the expression of K-ras between gastric cancer and precancerous lesions.2.K-ras is positively correlated with cell aging index.
