Objective:By detecting the distribution characteristics and laws of polymorphism of CAPN10gene,CDKAL1 gene,APOB gene and ZMIZ1 gene in Yan’an Han population,it is to investigate the correlation between the polymorphism of 6 SNP loci of these four genes and type 2 diabetes mellitus(T2MD)in Yan’an Han population.Methods:150 patients with T2MD diagnosed in Endocrinology Department of Yan’an University Affiliated Hospital from March 2017 to September 2018 and 100 healthy people examined in Physical Examination Center are selected.There is no significant difference in age and gender between the two groups.The genetic sequencing method is used to detect the polymorphism of SNP43 and SNP19 of gene CAPN10,rs7756992 and rs7754840 of gene CDKAL1,rs12571751 of gene ZMIZ1,and the polymorphism of rs679899 of APOB respectively.A genotyping database is established based on the detection data.SPSS 17.0 software is used to carry out statistical analysis,and t test and x~2 test were performed on the age difference and gender difference in the two groups of independent samples.The blood sugar levels of T2MD group and healthy control group are tested by x~2.Analysis whether the frequency of genotype distribution of the four genes in the type 2 diabetes group and the healthy control group conforms to the HW balance law.Statistical analysis is carried out on the differences of SNP43,SNP19,rs7756992,rs7754840,rs12571751,rs679899 genotype and allele frequency distribution between T2MD group and healthy control group.Results:1.Comparison of general data of subjectsThe age of T2DM group and healthy control group are 53.50±12.90 years old and50.99±11.29 years old respectively,with no significant difference between the two groups(t=1.583,p>0.05);There are 92 men and 58 women in the T2DM group and 57 men and43 women in healthy control group respectively,and there is no significant difference in the gender composition between the two groups(t=0.468,p>0.05).The levels of FBG in T2DM group and healthy control group were 11.05±4.31 mmol/L and 5.46±0.56 mmol/L respectively,and the difference was statistically significant(t=12.895,p<0.05).2.Hardy-Weinberg Equilibrium TestThe genotype frequency distribution of SNP43 of gene CAPN10 in the T2DM group and healthy control group does not conform to HW balance(p<0.05);The genotype frequency distribution of SNP19 of gene CAPN10 is consistent with HW balance(x~2=0.15,P=0.928;x~2=0.68,P=0.711)in T2DM group and healthy control group;The genotype frequency distribution of rs7756992 of gene CDKAL1 conforms to HW balance(x~2=2.48,P=0.289;x~2=0.53,P=0.766)in both T2DM group and healthy control group;The genotype frequency distribution of rs7754840 of gene CDKAL1 conforms to HW balance(x~2=5.86,P=0.053;x~2=0.04,P=0.978)in both T2DM group and healthy control group;The genotype frequency distribution of rs12571751 of gene ZMIZ1conforms to HW balance in T2DM and healthy control group(x~2=0.77,P=0.682;x~2=0.55,P=0.760);The genotype frequency distribution of rs679899 of gene APOB in T2DM group and the healthy control group conforms to HW balance(x~2=5.71,P=0.058;x~2=0.08,P=0.959).3.Detection results of SNP43 allele of gene CAPN10SNP43 allele is G homozygous in both T2DM group and healthy control group,the distribution frequency is 100%,and there is no significant difference in allele type and frequency(p>0.05).4.Detection results of polymorphism of SNP19 in gene CAPN10There is no significant difference in the frequency distribution of SNP19 genotype between T2DM group and healthy control group(x~2=2.459,P>0.05);There is no significant difference in the frequency distribution of SNP19 genotype between T2DM group and healthy control group(x~2=1.688,P>0.05).5.Detection results of polymorphism of rs7756992 allele of gene CDKAL1There is significant difference in the frequency distribution of rs7756992 between T2DM group and healthy control group(x~2=6.276,P<0.05).There is significant difference in the distribution frequency of G and A alleles between T2DM group and healthy control group(x~2=4.033,P<0.05).Individuals carrying G alleles(including homozygous and heterozygous)are 1.45 times more likely to develop T2DM than the individuals with AA homozygous.6.Detection results of polymorphism of rs7754840 allele of gene CDKAL1There is no significant difference in genotype frequency of rs7754840 between T2DM group and healthy control group(x~2=4.250,P=0.119).The frequencies of G and C alleles are not significantly different between T2DM group and healthy control group(x~2=1.466,P=0.226).7.Detection results of polymorphism of rs12571751 allele of gene ZMIZ1There is no significant difference in genotype frequency of rs12571751 between T2DM group and healthy control group(x~2=3.883,P=0.144).The frequencies of A and G alleles are not significantly different between T2DM group and healthy control group(x~2=2.581,P=0.108).8.Detection results of polymorphism of rs679899 allele of gene APOBThere is no significant difference in genotype frequency of rs679899 between T2DM group and healthy control group(x~2=6.686,P=0.035).The frequencies of A and G alleles have no significant difference between T2DM group and healthy control group(x~2=3.973,P=0.046).Individuals carrying A alleles(including homozygous and heterozygous)are 1.57 times more likely to develop T2DM than the individuals with GG homozygous.Conclusion:1.The polymorphism of rs7756992 of gene CDKAL1 and rs679899 of gene APOB polymorphism are correlated with T2DM in Yan’an Han population.2.Individuals carrying rs7756992 G allele of gene CDKAL1 have a high risk of T2DM.Individuals carrying rs679899 A allele of gene APOB have a high risk of T2DM.3.The occurrence of T2DM is associated with gene polymorphism.Detection of its polymorphism has certain value for preventing and delaying the onset of T2DM.
