Mappianthus tomentosus,a new species discovered from Guangxi,belongs to the genus Mappianthus of the Icacinaceae family.This genus consists of three species.We have studied the chemical constituents of M.iodoies,and found that it is rich in alkaloids and lignans.To further study the genus Mappianthus,in this paper we will study the chemistry constituents and anti-inflammatory activity of the M.tomentosus.The stems of M.tomentosus were investigated on their chemical constituents systematically by various kinds of chromatographic methods,33 compounds were obtained from EtOAc fraction,including 14 lignans(2,5–17),3 diphenyl ethers(1,3,4),2 triterpenes(18 and 19),2alkaloids(21 and 22)and 12 others(20,23–33).Their structures were identified by spectroscopic methods.These compounds were identified as follows:ethyl dichloroasterrate(1),mappilignan A(2),asterric acid(3),ethyl asterrate(4),balanophonin(5),erythro-1-(4-hydroxy-phenyl)-2-{4-[2-formyl-(E)-vinyl]-2-methoxy-phenoxy}-propane-1,3-diol(6),threo-1-(4-hydroxy-phenyl)-2-{4-[2-formyl-(E)-vinyl]-2-methoxy-phenoxy}-propane-1,3-diol(7),erythro-4,7,9,9?-tetrahydroxy-3,3?-dimethoxy-8-O-4?-neolignan(8),threo-4,7,9,9?-tetrahydroxy-3,3?-dimethoxy-8-O-4?-neolignan(9),γ?-methyl ether of guaiacylglycerol-β-coniferylether(10),coriarianeolignan A(11),tarennane(12),pinoresinol(13),8-hydroxypinoresinol(14),salicifoliol(15),lyciumin(16),chushizisin I(17),3β-O-cis-caffeoylbetulin(18),3β-O-trans-caffeoylbetulin(19),methyl 6-acetyl-4-methoxy-5,7,8-trihydroxynaphthalene-2-carboxylate(20),1-carbomethoxy-β-carboline(21),1H-indole-3-carboxaldehyde(22),4H-pyran-4-one,2-(ethoxymethyl)-3-hydroxy(23),ω-hydroxypropioguaiacone(24),4-hydroxy-3,5-dimethoxybenzaldehyde(25),vanillic acid(26),syringic acid(27),p-hydroxybenzaldehyde(28),3,4,5-trimethoxyphenol(29),3,4,5-trimethoxy benzy alcohol(30),2-(4-hydroxy-3,5-dimethoxyphenyl)ethanol(31),coniferyl aldehyde(32),trans-p-coumaryl aldehyde(33).The anti-inflammatory activity of compounds 1-4 and 17 was tested with NO production as an indicator by lipopolysaccharide(LPS)-induced BV-2 microglial cells.Only compound 2showed that inhibition to BV-2 microglial cells,with IC50 value of 38.87μM.
