The more somatic mutations accumulate in tumor cells,the higher the risk and possibility of recurrence is.Meanwhile,the higher the immune system reacts to the tumor cells,the more difficult the tumor cells grow and the tumor is less likely to relapse.Therefore,to better assess the risk status of cancer patients and characterize cancer patients' survival,we proposed to integrate tumor mutation burden(TMB),immunoreactivity and relative abundance of immune cells to construct a ratio model of immunity and tumor somatic mutation accumulation.Our study was focused on 6,341 samples of 16 cancer types in the Cancer Genome Atlas(TCGA),we constructed three models,including the ratio of immunoreactivity to TMB,the ratio of relative abundance of immune cells to TMB and a model of integrating immunoreactivity,relative abundance of immune cells and TMB.The related models were then used to score the risk status of cancer patients and explore the correlation between risk status and patient prognosis.The internal mechanisms of high-risk group and low-risk group were further explored through differential expression and gene function enrichment analysis.The results showed that for the 16 cancers,the “ratio of immunoreactivity to TMB model” could predict the disease free survival of 9 cancer types,but only predict the top 20% of patients and the last 20% patients;the model with "the ratio of relative abundance of immune cells to TMB" could predict the disease free survival of all patients for 7 cancer types;and "combined immunoreactivity,relative abundance of immune cells and TMB models" could predict disease free survival of 11 cancer patients.Taking liver hepatocellular carcinoma as an example,all three models can predict the disease free survival of patients(pvalue=0.00343,2.48e-05,2.30e-05).
