Influenza virus is highly contagious and spreads rapidly,posing a great threat to human health.At present,there is a lack of effective vaccine and antiviral drugs for prevention and treatment.In recent years,there has been increasing interest in the study of plant extracts and their derivatives to paly role in antivirus,which has great development significance.Garlic is a kind of edible and medicinal plant,containing a variety of bioactive substances.The most effective active ingredient is diallyl trisulfide.Relevant studies have shown that diallyl trisulfide has an effective effect in improving the body's immunity,anti-inflammation and anti-foreign pathogenic microorganisms.From the perspective of the pathogenesis of influenza,innate immunity is the first line of defense against influenza virus infection,in which RIG-I-like receptors play an important role in the natural immunity against influenza virus.However,how diallyl trisulfide plays an antiviral role and how to regulate the body's immune response has not been reported.Based on this,this study will investigate the effect of diallyl trisulfide on influenza virus in vivo and in vitro respectively,and the role of RIG-I in the natural immune signaling pathway.The main research results are as follows:1)Vitro Test: When A549 cells were infected with H9N2 avian influenza virus,diallyl trisulfide could significantly improve the activity of A549 cells and inhibit the proliferation of influenza virus in the different replication processes of drug pretreatment,co-treatment and post-treatment groups as well as virus particles.In addition,diallyl trisulfide can also down-regulate the expression of inflammatory factors induced by influenza virus at the m RNA level,and significantly up-regulate the expression of related antiviral genes in the RIG-I signaling pathway at the m RNA and protein levels.Finally,the RIG-I interference test further proves that diallyl trisulfide may play the role of anti-influenza virus through the RIG-I signaling pathway.2)Vivo Test: When BALB/c mice infected with H9N2 avian influenza virus,diallyl trisulfide can significantly improve the clinical symptoms of influenza virus infection in mice,reducing weight and pulmonary edema and pulmonary pathological changes in mice,such as pulmonary inflammatory cytokines IL-6,TNF-?,IL-10 and the influenza virus M gene expression.Meanwhile,antiviral gene RIG-I and the expression of IFN – ? was improved.In addition,serum biochemical indexes analysis showed that diallyl trisulfide may improve liver function,reduce blood lipid,improve renal function and improve antioxidant activity.The above results show that diallyl trisulfide has significant inhibitory effect on influenza virus in vivo and in vitro,inhibit the inflammatory response and up-regulate the expression of related antiviral genes in the RIG-I signaling pathway.This study can provide reference for the research and development of novel anti-influenza virus drugs.It also provides a scientific basis and mechanism for the study of the antiviral effect of diallyl trisulfide.
