As one of the Chinese medicine homologous food,Flos Sophora Immaturus is rich in resources,nutrients and active ingredients.This experiment took Flos Sophora Immaturus as the research object,and explored the separation and purification of its functional components,the evaluation of its bioactivity,and the development of functional products.The study provides theoretical and scientific basis for the further development,utilization and the promotion of added value of Flos Sophora Immaturus.The Flos Sophora Immaturus was extracted by ethanol using ultrasonic assisted.The ethanol extract was successively partitioned into n-hexane,ethyl acetate and n-butanol fractions according to the polarity of the organic solvent.A variety of techniques such as silica gel column chromatography were used to separate and purify the ethyl acetate extract and n-butanol extract.The eight separated compounds were identified by mass spectrometry and nuclear magnetic resonance,and they were identified as isorhamnetin(1),kaempferol(2),quercetin(3),carotenoid(4),rutin(5),narcisin(6),D-pinitol(7)and quercetin 3-O-?-D-glucopyranosyl-(1?2)-[?-L-rhamopyranosyl-(1?6)]-?-D-glucopyranoside(8).The MTT assay was used to evaluate the effect of functional components in Flos Sophora Immaturus in inhibiting the proliferation of tumor cells.The results showed that compounds 1,2 and 3 could inhibit the proliferation of HepG2 cells to varying degrees.The order of inhibition of tumor proliferation activity among the three was: Compound 2 > Compound 1 > Compound 3.After treating cells for 72 hours with compound 2(kaempferol),the inhibition rate reached 68.19% at the highest dose taken in the test range.The functional solid beverage was developed based on Flos Sophora Immaturus.Based on the results of single factor test,the extraction rate of total flavonoids was used as the response value,and the optimum process conditions were obtained by response surface methodology: the particle size of the material was 80 mesh,and the ethanol concentration was 70%,the solid-liquid ratio using ethanol was 1:20(m:v),the solid-liquid ratio using water was 1:40(m:v),the extraction time was 4 min,and the flash extraction voltage was 110 V.The extraction rate at this time was 25.81%.The Flos Sophora Immaturus tea powder rich in total flavones and jujube powder were combined.With sensory evaluation,the optimal ratio of the two kinds powder is 1:2(m:m).The amount of white sugar can be added according to individual taste and requirements,and boiled water is the best way to brew.
