Noni enzyme,a produce from the natural fermentation of noni fresh fruit,has a variety of health functions,and it's popular in the market.But up to now,few researches were focused on the chemical composition of noni enzyme and the impact of fermentation methods on its component content and activity.In order to deal with these problems,the chemical constituents of noni enzyme and its fermentation methods were studied in this paper.The contents are as following:1?Twenty-seven compounds were isolated from noni enzyme by various chromatographic techniques.They were xylogranatinin(1),pelargonic acid(2),1,5,15-Tri-O-methylmorindol(3),1,5,7-Trihydroxy-6-methoxy-2-methoxymethylanthraquinone(4),Sesquipinsapol B(5),(+)-syringaresinol(6),pinonesinol(7),3-benzyl-6-isobutyl-piperazine-2,5-dione(8),Bungein A(9),3-methylhexahydropyrrolo[1,2-a]pyrazine-1,4-dione(10),2(S)-3?-hydroxybutan-2?-yl-2-hydroxypropanoate(11),3-hydroxy-3-methyl-2-hydroxybutyl propionate(12),3-(sec-butyl)-6-methylpiperazine-2,5-dione(13),cyclo-(_L-Pro-_L-Leu)(14),vomifoliol(15),Cyclo-(Ala-Val)(16),lactic acid(17),scopoletin(18),3-(2-hydroxy-4,5-dimethoxyphenyl)propanoic acid(19),2-furoic acid(20),methyl-3-(2,4,5-trihydroxyphenyl)propanoate(21),p-hydroxy-benzoicacid(22),3-methoxy-4-hydroxy-benzoic acid(23),gentisic acid(24),medioresinol(25),hydroxychavicol(26),3?-hydroxybutan-2?-yl-2-hydroxypropanoate(27)?All the compounds,except 18?22?24,were obtained from noni enzyme for the first time.The inhibitory effect of compounds 1~8,10~15,17~27 on MGC-803 cells were studied,the result indicated that their has a lesser effect on MGC-803 cells.2?In this study,five noni enzymes were fermented using natural fermentation and the addition of strains including yeast,aspergillus niger,lactic acid bacteria,and red yeast element.According to the analysis of the chemical composition of the five noni enzymes,the contents of scopoletin,gentisic acid,caffeic acid,sinapic acid,quercetin,total phenol and total polysaccharide in natural fermentation noni enzyme were higher than other four enzymes.The contents of p-coumaric acid,ferulic acid,soluble protein,total flavonoids and the pH among the five enzymes were not significant difference.3?The evaluation results of antioxidant activity showed that,the total reduction ability and DPPH radical scavenging ability of Noni enzyme that prepared by natural fermentation was higher,and the hydroxyl radical scavenging ability and superoxide anion radical scavenging ability of five enzymes were less different.The correlation of component content and activity of noni enzyme was analyzed by SPSS,and the results showed that,the major antioxidants in noni enzyme were total polysaccharide and total phenol,which were mainly attributed to scopoletin,quercetin,gentisic acid and caffeic acid.In addition,studies also have found that noni enzyme has a certain tyrosinase inhibiting activity and inhibition ability to cell migration,but its activity size is not related with antioxidant activity.
