Amide bond is a very important functional group.It is ubiquitous in natural products,pharmaceuticals,and polymers.Recently,many amide compounds have been shown to be effective in the treatment of diseases such as cancer,Alzheimer's disease and hypertension.Therefore,the construction of the amide bond is of great significance.In this paper,the amide has been used as a substrate to oxidatively acylate with an aldehyde to construct a secondary amide.And this method has been applied to the synthesis of aniracetam.In addition,we also have studied the decarboxylated acylation reaction of?-oxocarboxylic acid.This paper includes the following four chapters.The first chapter reviews the recent progress in the oxidative acylation of primary amines,secondary amines,amides with aldehydes.The second chapter mainly studies the oxidative acylation of secondary picolinamides and aromatic aldehydes.Firstly,we have introduced the research significance of the subject.Then we have optimized the conditions of the reaction.The reaction constructs 20 amide compounds by oxidation acylation using Pd?OAc?2 as the catalyst,TBHP as an oxidant and TFA as an additive under 40oC in water.The substrate has good adaptability and the reaction yield is 45-87%.This method using water as a solvent makes the reaction to be more environment friendly.The reaction condition is mild and the aldehyde as an acylating agent also exhibits good atomic economy.The third chapter mainly researches synthetic process of aniracetam.Firstly,we review its synthetic routes.Then,the oxidative acylation system in the second chapter was applied to the synthesis of aniracetam.After the condition optimization the target product is successfully obtained?yield40%?.It provides a new method for the synthesis of aniracetam.In the fourth chapter,we have reviewed the decarboxylation coupling reaction of?-oxocarboxylic acid in recent years.Then,we research decarboxylation coupling reaction employing N-phenylmorpholine-4-carboxamides as substrate and?-oxocarboxylic acid as acylation.A superior reaction system has been found and substrate suitability has been studied?yield 42-62%?.The method has the advantages of mild reaction conditions and good atomic economy.
