Euonymus bungeanus belongs to the genus Euonymus of the family Celastraceae,it is a deciduous shrub or arbor.It is commonly used in gardens and green spacesin as ornamental tree.All plants of Euonymus bungeanus can be used as medicine with greater medicinal and economic value.In recent years,there was only a few studies on Euonymus bungeanus,.The study choosed Euonymus bungeanus fruit as the research subject and it was mainly about the extraction process,separation,purification and biological activities of its polysaccharides systematically,The aim of the study was to provid basic knowledge for the development of medicinal value of Euonymus bungeanus.1.The study used traditional hot water decoction method and ultrasonic assisted water extraction method to extract polysaccharide from Euonymus bungeanus fruit.The extraction conditions of two methods were optimized by the response surface method.The optimal extraction process was selected by using the yield of polysaccharide as the inspection index.The results showed that the best conditions of hot water extraction are:the ration of solid to liquid is 1:30,the water bath temperature is 90?,the time of extraction is 1.5 h,repeat 5times,the average yield is 10.9%,a 0.22%difference with the theoretical value.The best conditions of ultrasonic assisted water extraction method are:the ration of solid to liquid is 1:31,the water bath temperature is 49?,the time of extraction is 24 min,repeat 5 times,the average yield is 10.88%,a 0.53%difference with the theoretical value.2.For the results of the separation and purification of polysaccharides from Euonymus bungeanus fruit,protein impurities in the polysaccharides were removed by the Sevage method and polysaccharides were obtained by water extraction and alcohol precipitation The best conditions optimized by the response surface method are as follow:the ration of Chloroform to n-butanol is 4.5:1,the ration of EBFP to Sevage reagent is2.5:1,oscillation for 16.5 minutes,repeat 5 times,the average yield is 23.57%,a 0.9%difference with the theoretical value.3.The purity of the polysaccharide purified by D101 macroporous resin chromatography was 90.23%.Physicochemical properties results show that there are a large amount of reducing sugar in Euonymus bungeanus fruit.TLC results show that it may contain fructose,arabinose,galactose,glucose,glucuronic acid.DEAE-52 ion exchange column were used for Polysaccharide chromatography and five components EBFP-I,EBFP-II,EBFP-III,EBFP-IV,EBFP-V were eluted,and the contents of them were55.21%,3.01%,11.24%,16.2%,and 5.14%,respectively.4.The EBFP-I component was subjected to high performance liquid detection.The results showed that the monosaccharide composition contained glucose,fructose,and galactose.5.The extract of Euonymus bungeanus fruit polysaccharide has antioxidant activities,antibacterial effects and anti-tumor activities in vitro.The results showed that the polysaccharide EBFP had the highest scavenging ability to hydroxyl radical,up to 60.34%,and 53.1%at DPPH scavenging capacity,the scavenging ability of superoxide radicals was the weakest,with the highest being 42.34%.The scavenging ability of superoxide radicals was the weakest,with the highest being 42.34%.The antibacterial effect of EBFP polysaccharide from Euonymus bungeanus fruit showed good antibacterial effect on Escherichia coli and Staphylococcus aureus.The MIC values were E.coli 50 mg/mL and S.aureus 25 mg/mL.The cells in the experimental group showed obvious cell apoptosis.The IC50 of the SP2/0 cells was detected by the crystal violet method and the result was 37.753?g/mL,it had no significant effect on the BHK of normal cells.
