Evaluation Of The Value Of Alkaline Phosphatase And PIK3R1 In The Clinical Application Of Cervical Cancer

Objective:Explore the clinical application value of serum alkaline phosphatase(ALP)and PIK3R1 in cervical cancer(CC),to search for reasonable clinical indexes related to the development and progression of CC.Methods:This study was divided into two parts:The first part: Using the digitized case system of Affiliated Hospital of Jiangsu University,we retrospectively studied CC patients who received treatment from June2013 to June 2017.At the same time,relevant information was collected from healthy women in physical examination center of Jiangsu University.The values of serum CA-125,CA19-9,AFP,CEA,ALP,CHOL and TG were recorded respectively in CC patients and healthy group.The second part: Hematoxylin-Eosin(HE)staining was used to identify the morphological structure of CC.The levels of PIK3R1 gene in cervical squamous cell carcinoma(SCC)was detected by real-time quantitative PCR.The expression of p85?protein(encoded by PIK3R1 gene)in SCC tissues and benign cervix tissues was detected by immunohistochemistry(IHC).Results:(1)The serum CA-125,CA19-9,ALP,CHOL and TG values of CC patients were higher than those of healthy women,and all had statistical significance(P<0.05).The level of serum ALP in SCC at different FIGO stages were both higher than those in healthy women and all had statistical difference(P<0.01).The sensitivity and specificity of ALP in the diagnosis of SCC were 0.720 and 0.730 respectively.(2)Compared with normal cervical tissue,the expression of PIK3R1 in SCC was down regulated both of p85? protein(P<0.01).The expression level of p85? protein is related to the FIGO staging and metastasis of tumor.The later the tumor stage,the lower the expression level of p85? protein(P<0.01),the expression of p85? protein inmetastasis tumor tissue is lower than that of no metastasis(P<0.05).There was no significant difference in the expression level of p85? protein between age and degree of differentiation(P>0.05).(3)There is no correlation between serum ALP and PIK3R1 in SCC.Conclusion:(1)Serum ALP had a highly sensitivity and specificity in the diagnosis of SCC and closely related to the development of SCC.It can be used as an important indicator for the clinical evaluation of SCC.(2)PIK3R1 gene and its encoded protein p85? were significantly reduced in SCC suggesting that PIK3R1 play key roles of tumor suppressor in SCC.(3)There is no correlation between serum ALP and PIK3R1.
