Objective: To find out UNC5 C truncated mutation’s influences on the survival function of the Renal cancer cells.Methods: Construct the stable-expression renal cancer cell lines of the wild-type and the mutation-type by lentiviral infection,a series of cell function experiments and examinations on apoptosis markers are carried out afterwards.Co-transfection of the wild-type and the mutation-type to figure out this mutation’s effect on the wild-type.Results: Through the public database,we identified that the expression of UNC5 C in renal cancer tissues is more than that in normal ones.In addition,we found patients with high expression of UNC5 C have better prognosis.We found 14 cases have mutation in the N0.491 amino acid on the UNC5C’s protein sequence,which are identified as truncated mutation.We construct plasmid for this mutation and its wild type for stable expression cell lines.Through a series of experiments,we found that the mutation of UNC5 C can stabilize the protein expression of UNC5 C simultaneously inducing the expression of its ligand NTN1,thus promote the survival function of renal cancer cells.Conclusion: We found Q491Pfs*3 mutation of UNC5 C can stabilize the protein expression of UNC5 C,simultaneously inducing the expression of its ligand NTN1,thus promote the survival function of renal cancer cells.The biological effects of the mutation of the dependence receptor UNC5 C were further understood through experiments,which provided ideas for the study of other mutation sites.
