Objective:Primary liver cancer(PLC)is one of the common malignant tumors of the digestive system in China.7-ethylcamptothecin is a novel camptothecin derivative in which the 7-position of the B ring is substituted by ethyl.Camptothecin and its derivatives can target to inhibit topoisomerase I and show great antitumor effect in clinical.This study was to investigate the antitumor effect of 7-ethylcamptothecin in hepatocellular carcinoma cells and further explore its mechanism of action.Methods:CCK-8 assay was used to detect the proliferation ability on HCC cells.Next,the effect on the clonal formation of HCC cells were evaluated by clone formation assay.Cell cycle kit,cell apoptosis kit and Hoechst33342 were used to detect cell cycle and apoptosis of HCC cells.Then,the expression of caspase-3 and its substrate PARP protein in HCC cells were analyzed by Western blotting.The same method was used to detect on the expression levels of caspase-9 and Bcl-2 family proteins.Results:7-ethylcamptothecin suppresses human HCC cell proliferation and colony formation(P <0.01).7-ethylcamptothecin induced HCC cell cycle arrest in G1 phase(P <0.01).7-ethylcamptothecin induces HCC cells apoptosis in a caspase-dependent manner(P <0.01).7-ethylcamptothecin increases the activity of the apoptotic protein caspase-3 and its substrate PARP.Finally,7-ethylcamptothecin induces HCC cells apoptosis through a mitochondria-associated apoptotic pathway.Conclusion : 7-ethylcamptothecin inhabits proliferation and induces cell apoptosis which exerts a potent effect on human HCC cells.And 7-ethylcamptothecin induces HCC cells apoptosis through mitochondrial apoptosis pathway.
