Objects: In this study,we investigated the effects of Fruit of Ligustrum Obtusifolium Extract(FLOE)on the proliferation,migration and expression of apoptosis-inducing factor(AIF)in colon cancer HCT-15 cells in vitro,providing more theoretical basis for the development of traditional Chinese medicine in the treatment of colon cancer and the application of AIF in colon cancer therapy.Methods: Colon cancer HCT-15 cell was cultured in vitro,and logarithmic growth phase cells were used for experiments.The FLOE was diluted to different concentrations with RPMI-1640 incomplete medium and grouped into blank control group(0?g/ml)?2.5?g/ml?5?g/ml?10?g/ml?20?g/ml group and 75% alcohol was diluted with RPMI-1640 incomplete medium into negative control group(20?g/ml).The morphological changes of cells in different concentration groups were observed by ordinary light microscopy after 24 h,48h,72 h,96h and the number of adherent cells was calculated under microscope.MTS method was used to detect different concentration groups of FLOE on the effects of the cell viability before and after treatment for 24h?48h?72h and 96 h.Colony experiment and scratch test was used to detect the effect of FLOE before and after treatment on the proliferation and migration ability of HCT-15 cells in different concentrations.The expression levels of Survivin?Mcl-1 and AIF were detected by Western blot.Results: With the increase of FE concentration and prolonged action time,HCT-15 cells gradually deteriorated,cell gap gradually increased,cell vacuoled,decellularized cells increased and cell debris increased under microscope.The results of MTS showed that the cell viability after FE treatment was significantly lower than the control group(P<0.05).The cell viability was within the scope of the study and the drug effect.Concentration and duration of action were negatively correlated.The results of scratch test showed that the scratch area relative width after FLOE treatment was significantly wider than the control group(P<0.05).The colony experiment showed that the number of cell were significantly lower than control group(P<0.05).The results of Western bolt showed that the expression of Survivin ? Mcl-1 reduced and AIF increased with the increase of FLOE concentration.Conclusions: Fruit of ligustrum obtusifolium extract inhibited the proliferation and migration of HCT-15 cells significantly.The inhibitory effect was positively correlated with FLOE concentration and duration of action.Its mechanism may be achieved by down-regulating the expression of anti-apoptotic proteins and up-regulating the expression of apoptosis-inducing factor AIF.
