Objective:To observe the expression of ?3?1 integrin and the absolute number and relative density of podocytes in patients with hepatitis B virus associated glomerulonephritis(HBV-GN),and to investigate the changes of glomerular podocyte number and ?3?1 integrin in hepatitis B.The role of viral-associated glomerulonephritis(HBV-GN),and the correlation between the absolute number and relative density of podocytes and clinical indicators.Methods: Twenty-two patients with HBV-GN were enrolled in the study.Thirty patients with renal tumors were used as control group.Immunohistochemical staining of podocytes was performed with WT1 monoclonal antibody.Single glomerular podocytes were estimated by manual counting and pathological image analysis.Absolute number,the relative density of podocytes in a single glomerulus was determined.Glomerular immunohistochemical staining was performed using ?3 polyclonal antibody and ?1 polyclonal antibody to determine the average optical density value of glomerular ?3?1 integrin.Results: The absolute number and relative density of glomerular podocytes in HBV-GN group were lower than those in the control group,and the level of ?3?1 integrin was decreased compared with the control group.The absolute number of podocytes was negatively correlated with 24-hour urine protein quantitation(P<0.05),and no significant correlation was found with creatinine(P>0.05).?3?1 integrin was positively correlated with the absolute number and relative density of podocytes(P<0.05).Conclusion: Decreased expression of glomerular ?3?1 integrin in HBV-GN group may cause podocyte adhesion function to decline,resulting in podocyte shedding,which causes the number of glomerular podocytes to decrease,resulting in proteinuria in patients.These changes may be related to the pathogenesis and progression of hepatitis B virus-associated nephropathy.
