Objective: Neublastoma(NB)is an embryonic malignant tumor that is easy to metastasize in early childhood and affects the normal development of adrenal medulla and paraspinal sympathetic ganglion.According to previous reviews and studies,some molecular biological characteristics of human neuroblastoma have been found,mainly covering abnormal genomic and transcriptome level studies.Neuroblastoma can occur almost anywhere in the sympathetic nervous system.Data from recently published international cohort studies of large patients show that neuroblastoma often occurs in the adrenal gland(47%),peritoneal/retroperitoneal region(24%),neck(3%),chest(15%),pelvis(3%)and other areas(8%).Ginsenoside Rh2(ginsenoside Rh2),as a key active ingredient in ginseng,has attracted extensive attention in recent years.It has been found that ginsenoside Rh2 has strong anti-tumor activity in many human tumors.At present,related studies at home and abroad have found that ginsenoside Rh2 can inhibit the proliferation,migration,invasion of tumor cells,induce apoptosis and its role in tumor microenvironment.The application of ginsenoside Rh2 in neuroblastoma has not been reported.Therefore,the effects of ginsenoside Rh2 on the proliferation,apoptosis,migration and invasion of neuroblastoma were studied.Methods: Ginsenoside Rh2(0,5,10,20,40,80?g/ml)was used to interfere with neuroblastoma cell line SH-SY5 Y.Cell counting kit-8(CCK-8)method was used to detect the effects of ginsenoside Rh2 on the proliferation of neuroblastoma SH-SY5 Y.Cell scratch test and Transwell chamber model were used to detect the migration and invasion of neuroblastoma SH-SY5 Y treated with ginsenoside Rh2.Western-bolt(WB)test was used to detect neuroblastoma SH-SY5 Y.Expression of matrix metalloproteinase 2(MMP2),Bax,Bcl-2 and ?-catenin proteins in SH-SY5 Y cells.Result: Ginsenoside Rh2 could inhibit the proliferation of neuroblastoma in a time-concentration dependent manner.In CCK-8 results,24 hours showed a slight cytotoxicity,48 hour performance for toxicity.Ginsenoside Rh2 could inhibit the migration and invasion of neuroblastoma.Western blotting showed that ginsenoside Rh2 decreased the expression of migration-related protein MMP2,increased the expression of apoptotic protein Bax and decreased the expression of anti-apoptotic protein Bcl-2.The level of ?-catenin in neuroblastoma treated with ginsenoside Rh2 decreased.Conclusion: 1.Ginsenoside Rh2 can inhibit the proliferation of neuroblastoma SH-SY5 Y in vitro,induce its apoptosis,inhibit its migration and invasion;2.Ginsenoside Rh2 down-regulates MMP2 protein to inhibit the migration and invasion of tumor cells,and inhibits the growth of neuroblastoma cells through ?-catenin protein in the Wnt pathway.
