Objective:Liver cancer is a common malignant tumor of digestive system.In our country,the main patnogenesis of liver cancer is viral hepatitis.The median age of patients with liver cancer in China is 40 ~ 50 years old,male is more than female,the incidence of malignant tumor is the third,and the mortality of tumor is the second.It has mortality is high and the prognosis is poor.The pathogenesis of hepatocellular carcinoma is very complex.The current treatment is mainly surgical treatment,supplemented by systemic radiotherapy,chemotherapy and minimally invasive interventional therapy However,the recurrence rate and metastasis rate are still high after treatment.Tumor microenvironment is the local internal environment of tumor cell growth,including a variety of cells,cytokines and extracellular matrix,and so on.The occurrence and development of tumor is closely related to tumor microenvironment.For a long time,people pay more attention to the study of tumor itself,and often ignore the tumor microenvironment.In recent years,more and more studies have found that tumor microenvironment plays a very important role in the occurrence and development of tumor.Tumor immunotherapy is a new treatment after the traditional therapy.It is a method of applying the principle of immunology to regulate the immune system of the body in an active and passive way,so as to achieve the anti-tumor effect.In 2013,the American Journal of?Science?ranked tumor immunotherapy as the top ten technological breakthroughs in the world.IL-34 is a novel cytokine discovered in 2008,which mediates its biological function by binding to M-CSFR,a receptor expressed on the surface of monocytes.It can promote the proliferation activity of monocytes and the formation of macrophage colony forming units in bone marrow culture in vitro.It can promote the infiltration of TAMs and regulate the growth,infiltration and metastasis of tumor.In this study,we analyzed the difference of IL-34 expression between tumor tissue and paracancerous tissue in patients with HCC and the correlation with clinicopathological features of HCC.To explore the role of IL-34 in the microenvironment of hepatocellular carcinoma(HCC),and to provide some objective evidence for the treatment of HCC,especially immunotherapy.Methods:1.From January 2018 to August 2018,43 cases of hepatocellular carcinoma were collected from the first affiliated Hospital of China Medical University.2.Expression of IL-34 in hepatocellular carcinoma tissues and adjacent tissues of 43 patients with hepatocellular carcinoma by immunohistochemical method 3.According to its expression,it was divided into high expression group and low expression group.The intergrate optical density value(IOD)was analyzed by Image-pro plus 6.0 software.4.To determine the relative expression of IL-34 in tumor tissues and adjacent tissues,and to explore the relationship between IL-34 expression and age,sex,tumor size,liver cirrhosis,tumor differentiation,HBV copies,HBs Ag,in paracancerous tissues.Relationship between clinical and pathological features such as portal vein tumor thrombus and serum AFP,TNM staging.Results:Immunohistochemical results showed that the expression of IL-34 was significantly higher in paracancerous tissues than in tumor tissues(p<0.05),There was a correlation between the size of IL-34 expression and tumor size in paracancerous tissues(p<0.05).There was no correlation between IL-34 expression and age,sex,liver cirrhosis,tumor differentiation,hepatitis B copies,portal vein tumor thrombus,serum AFP and TNM stage in paracancerous tissues(p>0.05).Conclusion:The expression of IL-34 in HCC adjacent tissues was higher than that in tumor tissues,and the expression of IL-34 in paracancerous tissues was correlated with tumor size,suggesting that IL-34 might be involved in the pathogenesis and development of liver cancer,It can provide some objective basis for the treatment of liver cancer.The expression of IL-34 was not correlated with age,sex,liver cirrhosis,tumor differentiation,DNA copies of hepatitis B,HBs Ag,portal vein tumor thrombus,serum AFP and TNM stage,etc.
