Objective To investigate the expression of ?-smooth muscle actin(?-SMA)and E-cadherin(E-cadherin)in renal tissue of renal interstitial fibrosis in mice,and to investigate the effect of breviscapine on renal interstitial fibrosis.Reverse transformation.Methods A total of 72 male CD1 mice of 4 weeks old were selected as the study subjects.They were randomly divided into sham operation group,renal interstitial fibrosis rat model group with unilateral ureteral obstruction,and breviscapine group.A total of 24.Among them,the breviscapine group was given intraperitoneal injection of breviscapine injection(2.5 ml/time,twice/d)from 1 day before the operation to the day of the operation,while the sham operation group and the renal interstitial fibrosis group were both.In the same period,the same volume of normal saline was intraperitoneally injected.The mice were sacrificed at 4,7 and 14 days after operation,and 8 mice were sacrificed.The left kidney was removed and the kidney was cut into two parts.Histopathological sections and immunohistochemistry were detected,and another part was frozen in liquid nitrogen for Western blotting.The renal tissues of each group were collected to observe the pathological changes of renal tissues in each group.The serum creatinine and urea nitrogen levels of each group were detected.Immunohistochemistry and Western blot were used to detect the expression of ?-SMA and E-Cad proteins in the kidney.Results The serum creatinine and urea nitrogen levels of the three groups of mice were compared.The results showed that the BUN and Scr levels in the model group were significantly higher than those in the sham operation group,and the difference was statistically significant(p<0.05).The BUN and Scr levels in the flowering group were significantly lower than those in the model group(p<0.05).At 14 days after operation,tubulointerstitial lesions continued to worsen,renal interstitial inflammatory cells were still diffusely infiltrated,renal tubules were significantly dilated,and renal interstitial fibrosis was evident.Comparing the relativeareas of renal interstitial between groups,suggesting scutellaria Compared with the model group in the same period,the relative area of ??renal interstitial was significantly decreased,and the difference was statistically significant(p<0.05).The results of immunohistochemistry and Westernblot showed that the expression of?-SMA at 4,7 and 14 days after breviscapine group was significantly lower than that of the model group at the same time point(p<0.05).The expression of E-Cad was significantly decreased in the model group at 4 days after operation,and decreased significantly with the prolongation of obstruction.The expression of E-Cad A at 4,7 and14 days after breviscapine group was significantly higher than that at the same time.Group(p<0.05).(See Table 2).Conclusion Breviscapine has anti-renal interstitial fibrosis;breviscapine can reduce renal interstitial fibrosis and delay the progression of renal failure in mice by down-regulating ?-SMA and increasing E-Cad;breviscapine can inhibit The process of tubulointerstitial transdifferentiation(EMT),which in turn inhibits renal interstitial fibrosis.
