Bisphenol S(BPS),a structural analog of Bisphenol A(BPA),has been widely used as a substitute for epoxy resin,baby bottle,can liner,thermal paper,food packaging materials,and other products due to the limited application of BPA.BPS and BPA are both typical exogenous endocrine disruptors with estrogenic and antiandrogenic properties,which can interfere with the endocrine system and have a negative impact on the health of the body.In vivo and in vitro studies have indicated that BPA can break the balance of lipid metabolism and inducing lipid deposition act as obesogen.At present,there are few studies on the effects of BPS on lipid metabolism,and its mechanism of action is unclear until now.In this thesis,BPS was taken as the research object and C.elegans as the model organism to study the effects of BPS on lipid metabolism.Firstly,acute exposure was performed to study the toxic effect of BPS on C.elegans.Secondly,on this basis,the effects of BPS on the lipid metabolism and mechanism of C.elegans was studied.Finally,the effects of BPS on lipid metabolism and mechanism of C.elegans was studied by multi-generational exposure on the basis of the first generation.The main research contents and results are as follows:(1)The acute toxic effects of BPS on C.elegans were studied.The effects of BPS on LC50,locomotion behavior,body length,reproduction,lifespan,reactive oxygen species(ROS),and superoxide dismutase(SOD)levels of C.elegans were studied.The results showed that the LC50 value of BPS for C.elegans was 545.59 mg/L.When the concentration of BPS increased to 0.01μM,it can have a negative impact on C.elegans,which mainly include inhibition of locomotion behavior,shortening of body length,decrease of reproduction,reduction of life span,increase of ROS level,and decrease of SOD level.0.01μM BPS also could significantly inhibit the locomotion behavior,body length,and reproduction of C.elegans by multi-generational exposure.(2)The effects of BPS on lipid metabolism of C.elegans was studied.The results showed that Oil-Red-O staining,triglyceride,and free fatty acid were significantly increased,indicating that BPS could significantly induce lipid deposition of C.elegans.The effects of BPS on the lipid metabolism of C.elegans showed a non-monotonically increasing relationship,and 0.01μM BPS could affect the lipid metabolism of C.elegans.BPS may regulate nematode lipid metabolism by up-regulating daf-16,fat-5,fat-6,fat-7,and down-regulating nhr-49 and acs-2.BPS may affect the lipid metabolism of C.elegans through nhr-49 mediated nuclear hormone signaling pathway.(3)The effects of BPS on lipid metabolism of C.elegans was studied by multi-generational exposure.The results showed that the Oil-Red-O staining and triglyceride of C.elegans were still significantly increased through multi-generational exposure.BPS could still significantly induce the lipid deposition of the four generations of C.elegans.BPS may regulate lipid metabolism of C.elegans by up-regulating daf-16,fat-7,and down-regulating nhr-49 and acs-2 in multi-generational exposure.BPS can still affect the lipid metabolism of C.elegans through the nhr-49 mediated nuclear hormone signaling pathway in multi-generational exposure.
