| Objective:In the current study,we investigated the relationship between the level of ATⅢ and contrast induced nephropathy(CIN)in patients.We also examined therapeutic effect of ATⅢ on CIN in Sprague-Dawley rats.Furthermore,the potential mechanisms of this phenomenon are explored in several different ways.Methods:(A)Clinical research From July 1st 2014 to December 31 th 2015,we collected the data from patients with coronary angiography.The data includes information like gender,age,ATⅢ activity,and so on.Blood samples were collected 48 hours after sugery.(B)Vitro experiments24 healthy male Sprague-Dawley(SD)rats were randomly divided into four groups.Four groups are control group,contrast agent nephropathy group,preoperative injection of ATⅢ contrast agent nephropathy group,and postoperative injection of ATⅢ contrast agent nephropathy group.Control group and contrast agent nephropathy group were given vehicle.Results:1.The incidence of AKI was obviously higher in patients with low ATⅢ activity.2.Antithrombin Ⅲ attenuated contrast induced kidney impairment and kidney histological Injury.Administration of ATⅢ significantly attenuated the elevation ofserum creatinine(SCr),blood urea nitrogen(BUN)in the SD rat CIN model.3.Antithrombin Ⅲ attenuated kidney Inflammation and oxidative stress in CIN Rats.4.Antithrombin Ⅲ treatment effectively improved renal blood flow.5.Antithrombin Ⅲ had a significant effect on kidney apoptotic cells.Conclusions:Patients with low ATⅢ activity presented a higher incidence of contrast induced nephropathy following coronary angiography.Our data demonstrated ATⅢ significantly attenuated the elevation of SCr,BUN,and renal histological injury.ATⅢ seems to attenuate CIN by inhibiting inflammation,oxidative stress,apoptosis and improving renal blood flow. |
