Objective:Melanoma is a kind of skin tumor which originate from melanocyte.It has a median survival time of 6 months and a 5-year survival rate of less than 5%,which is most hazardous malignancy of skin cancer.Cancer stem cells are capable of self-renew and differentiation and have the potential for indefinite proliferation,a function throu--gh which they may cause tumor growth.In the present study,we aimed to investigate the expression pattern and clinical significance of three CSC markers,CD133,CD271 and ABCB5 in melanoma,and to correlate these with survival data.Methods:120 patients in Shanghai Sixth Peolpe Hospital was collected,in which include31 cases of cutaneous melanoma,29 cases of mucosal melanoma,28 cases of metast--atic melanoma and 32 cases of nevi.An immunohistochemical study for CD133 and CD271,ABCB5 was performed in 120 specimens to evaluate possible differences.cli--nicopathological varibles of cutaneous melanoma were evaluated retrospectively,to compare the results with survival data.Data were assessed using univariate and multi--variate statistics.Results:The highest IHS of CD133 was in metastatic melanoma and the lowest was in nevi.There was no difference between cutaneous melanoma and mucosal melanoma.The expression of CD271 in all lesions were low.The highest level expression is in m--etastatic melanoma.There was no difference in ABCB5 expression.On univariate an--alysis,CD133 and ABCB5 were statistically associated with the overall survival.On multivariate analysis,ABCB5 was an independent prognostic predictor for overall sur--vival.Conclusion:CD133 and CD271,which are the marker of cancer stem cells,may play an important role in the course of banal lesion-primary tumor-metastatic lesion in melan--oma.CD133 and ABCB5 were statistically associated with the overall survival.ABC B5 was an independent prognostic predictor for overall survival.
