Objective: To investigate the effect of 18 beta-sodium glycyrrhetinic acid on the expression of Nuclear Factor-kappaBp50(NF-κBp50)in nasal mucosa and Interleukin-4(IL-4)in serum of rats with allergic rhinitis(AR),and the correlation between NF-κBp50 and IL-4,then explore its significance.Methods: 100 wiatar rats were randomly divided into normal control group,AR model group,budesonide group and 18β-SGA low dose(20mg/kg)and high dose(40mg/kg)intervention groups after successfully establishing the model,with 20 rats in each group.Nasal mucosa tissue and serum were respectively taken after intervention for 2 weeks and 4 weeks.Immunohistochemical was used to observe the distribution of NF-κBp50 in nasal mucosa,western blotting and real-time qPolymerase Chain Reaction were used to detect the expression of NF-κBp50 protein and mRNA.The content of IL-4 in serum was detected by enzyme-linked immunosorbent assay.Results: 1.Immunohistochemistry showed that NF-κBp50 was mainly expressed in the cytoplasm and nucleus of nasal mucosa pseudostratified ciliated columnar epithelial cells,lamina propria glandular cells,vascular endothelial cells and inflammatory cells.2.Compared with the normal group,the expression level of NF-κBp50 and IL-4 in AR model group expressed more higher(P < 0.05).The expression in each treatment group was lower than that in AR model group(P < 0.05).3.Pearson correlation analysis showed that the expression of IL-4 in every group was positively correlated with the expression of NF-κBp50 protein and mRNA after intervention for 2 weeks and 4 weeks,P< 0.001,and the correlation coefficient was 0.792、0.790、0.930 and0.952,respectively.Conclusion: 1.NF-κB was overactivated and its expression was enhanced,the content of IL-4 in serum was significantly increased in allergic rhinitis rats.2.Pearson correlation analysis showed that the changes in NF-κBp50 protein andmRNA levels are strongly positive correlated with IL-4 expression.3.18β-SGA can play an anti-inflammatory role by inhibiting the activity of NF-κB signaling pathway,reduce the expression of p50 subunit in the nasal mucosa and the content of T helper cell-2 cytokine IL-4,which can alleviate the inflammatory response in the nasal mocosa of rats.4.Intervention with 18β-SGA 20mg/kg for 2 weeks was effective in the treatment of allergic rhinitis in rats and the absorption effect of nasal mucosa was better than that of 40mg/kg.
