Objective:Intermittent hypoxia mimics chronic obstructive sleep apnea hypopnea syndrome(OSAHS),investigates the relationship between intermittent hypoxia and renal injury and its association with Beclin1 mRNA expression,and studies the intervention of losartan.Provide new research ideas and theoretical basis for clinical prevention and treatment of OSAHS-related kidney diseases.Methods:To establish an intermittent hypoxic environment,60 SD rats were divided into 4 groups: group A(normal control group),group B(intermittent hypoxia group),and group C(intermittent hypoxia + losartan).Groups,group D(intermittent hypoxia + saline group),15 rats in each group,rats in group B,C,and D were placed in intermittent hypoxic environment,and rats in group A were placed in normal indoor air.In the intermittent hypoxia group,8 hours a day,continuous modeling for 6 weeks.At the end of modeling,6 rats were randomly selected,arterial blood was taken from the abdominal aorta,and kidney samples were collected.Blood samples were used to detect serum creatinine and urea nitrogen levels.Rat kidney samples were stained with hematoxylin-eosin.The pathological changes of rat kidney were observed under microscope.The ultrastructure of kidney was observed under electron microscope.The expression of Beclin1 mRNA in kidney tissue was detected by PCR.Results:(1)There was no significant difference between the blood urea nitrogen levels of the rats in the intermittent hypoxia environment and the normal control group(P>0.05);(2)compared with the normal control group,intermittent hypoxia group and intermittent hypoxia The serum creatinine level of the rats in the group + saline group was increased(P<0.05),which was statistically significant.Compared with the normal control group,the serum creatinine level in the intermittent hypoxic carolartan group did not change significantly(P >0.05);(3)Observing the pathological structure of kidney tissue in rats under electron microscope,it was found that some of the renal tissue of group C showed edema of renal tubules compared with group A,while kidney tissues of group B and group D Significant tubular edema,structural disorder,and stenosis were observed.(4)Compared with group A,the expression levels of Beclin1 mRNA in group B and group D were significantly increased(both P<0.05).Group B was better than group C.The expression level of Beclin1 mRNA was significantly increased(P<0.05).There was a difference between the expression levels of Beclin1 mRNA in the kidney tissues of group C and group D(P<0.05).There was no difference in the mean value of Beclin1 mRNA expression between the two groups(P>0.05).Conclusion:1.The establishment of intermittent hypoxia model was successful;2.Autophagy mediates the injury of rat kidney by intermittent hypoxia;3.Losartan reduces the damage of rat kidney by intermittent hypoxia,possibly by reducing excessive The occurrence of activated autophagy.
